摘要
目的:探讨人外周血树突状细胞(DC)的诱导培养及冻存方法和意义。方法:通过树突状细胞的分离和培养、抗原标本前处理、抗原的装载、实验分组、细胞冻存等,摸索出树突状细胞诱导扩增以及促进成熟的优化实验方案。结果:外周血单个核细胞可作为DC的来源,GM-CSF和IL-4可诱导其前体细胞向DC分化,添加促成熟细胞因子可以让细胞维持细胞生物活性。结论:人外周血DC细胞的诱导培养及冻存具有重要意义。冷冻DC细胞保持了它的特性,但会因冻融过程生物特性有所减低,为此在应用冻存DC细胞时应考虑增加输入细胞量。
Objective:To explore the significance and approach of inducing culture and cryopreservation in human peripheral blood dendritic cells. Methods : We fond the better experimental project of maturing and inducing the increase of dendritic cells by isolating and culturing , processing and loading antigen, classing cryopreserving cells, et al. Results : Dendritic cells derived from peripherial blood monocyte, after they were thawed and obtained bioactivity adding GM -CSF, IL-4. Conclusion: There is significant implication in inducing culture and cryopreservation in human peripheral blood dendritic cells.
