摘要
目的:探讨胃宁颗粒对人胃癌裸鼠原位移植瘤生长及转移的抑制作用。方法:制作人胃癌裸鼠原位移植模型,随机分成对照组、环磷酰胺(CTX)组、胃宁颗粒组、CTX+胃宁颗粒组,每组12只,观察瘤体积抑制率、瘤重量抑制率、肝转移发生率、肿瘤组织细胞形态学变化、凋亡指数(AI)、p53、Bcl-2、bax、CD44v6、增殖细胞核抗原(PCNA)蛋白表达的变化。结果:CTX组、胃宁颗粒组、胃宁颗粒组+CTX组和对照组相比,瘤重抑制率、肿瘤体积抑制率差异有显著性(P<0.05);胃宁颗粒组+CTX组与CTX组相比,瘤重抑制率、肿瘤体积抑制率差异有显著性(P<0.05);胃宁颗粒组、CTX组肝转移发生率与对照组相比,差异有统计学意义(P<0.05或P<0.01);胃宁颗粒组与对照组AI值相比,差异有显著性(P<0.01);胃宁颗粒组p53、bcl-2、bax、CD44v6、PCNA与对照组相比,差异有统计学意义(P<0.05);胃宁颗粒+CTX组与CTX组相比,p53、CD44v6、PCNA表达差异有显著性(P<0.05)。结论:胃宁颗粒具有抑制人胃癌裸鼠原位移植瘤生长及转移作用。
Objective:To investgate the inhibiting effect of Weining granule on the growth and metastasis of orthotopic human gastric carcinoma. Methods:To establish the model of orthotopic human gastric carcinoma,60 rats with orthotopic human gastric carcinoma were randomly divided into the control group, CTX group,Weining granule group,CTX with Weining granule group, 12 in each group. The inhibiting rate of carcinoma's volume and weight,the change of shape on carcinoma cells,the expression of apoptotic index (AI),p53,Bcl-2, bax, CD44v6 and PCNA were all measured. Results:The inhibiting rate of carcinoma's volume and weight in all CTX group,Weining granule group,CTX with Weining granule group were different from that of the control group(P 〈 0.05) ; the inhibiting rate of carcinoma's volume and weight in CTX with Weining granule group were different from that of the CTX group (P 〈 0.05) ; the rate of hepatic metastasis in CTX group and Weining granule group were less than that of the control group (P 〈 0.05, P 〈 0.01 ) ; AI, p53, bcl-2, bax, CD44v6 and PCNA expression in Weining granule group were significantly different from that in the control group (P 〈 0.05);p53, CD44v6 and PCNA expression in CTX with Weining granule group were less than that in the CTX group (P 〈 0.05). Conclusion:Weining granule can inhibit the growth and metastasis of orthotopic human gastric carcinoma.
