Analysis of Gamma-hydroxybutyric Acid using Gas Chromatography/Mass Spectrometry after Pentafluorobenzyl Derivatization

摘要 采用五氟卞基溴作为γ-羟基丁酸(GHB)的烷基化试剂,衍生化后进行了产物的净化处理,以降低衍生化产物对仪器的不利影响。考察了该衍生化反应的一些影响因素,优化了衍生化条件,并对衍生化产物进行了质谱解析。在此基础上考察了该方法的定量分析参数,结果表明,GHB在0.1~5mg/L的范围内,其衍生化产物具有良好的线性关系。在GHB浓度为1mg/L时,衍生化产物的相对标准偏差为13.4%。对添加尿样提取后进行衍生化并测试,检出限为10mg/L,与GHB的检测阈值相应。该研究建立了GHB分析的又一衍生化GC方法,可用于饮料中及生物样品中GHB的分析。 The γ-hydroxybutyric acid (GHB) was derivatized with pentafluorobenzyl bromide and then the product was purified to eliminate the disadvantageous effect. Several factors that affected the derivatization were examined and optimized. The GHB derivatives was analysed by MS. The linear relationship was in the range of 0. 1 -5 mg/L GHB, and the relative standard deviation was 13.4% at 1 mg/L level of GHB. The limit of detection (LOD) for spiked urine sample was 10 mg/L, at the lever of cutoff of GHB in urine. The method can be used in analysis of GHB in beverage and biological samples.
