聚合酶链反应检测粪便中的副溶血性弧菌的TDH基因

摘要 副溶血性弧菌是细菌性腹泻的主要致病因子.本研究建立一种快速标本处理和聚合酶链反应(PCR)诊断方法,扩增副溶血性弧菌的热稳定直接溶血素(TDH)基因。对一组56例腹泻患者粪便标本进行了检测,经细菌培养证实为副溶血性弧菌的9份标本,PCR法均阳性;培养法明性的39例中,有7例检测到TDH基因.统计学表明两种方法检测结果相当一致,但检出能力PCR法显著高于培养法。本法简便、快速、特异,适宜临床应用. Vibrio Parahaemolyticus is the principal pathogenic factor causing acute bacterial diarrhea. A method using the rapid PCR to detect the virulence gene, the thermostable direct hemolysin (TDH)of Vibrio para-haemolyticus, in diarrheal stools was set up. In 56 cases of diarrhea, 9 V. parahaemolyticus isolated by con-ventional culture method were positive with PCR; and TDH genes of 7 in 39 culture-negative samples were identified by PCR in feces. Statistical P values of both correlation and difference were less than 0.01 by Chisquare test. The results f the two methods are identical and PCR was more sensitive than cnventinalmethod. It is shown that the novel method is rapid, sensitive and specific for diagnosis of V. parahaemolyt-icus, and suitable for clinical use.
