秋水仙碱磁微球靶向治疗乳腺癌的实验研究 被引量:9

Orientation of Colchicine Magnetic Microsphere in Breast Cancer

摘要 目的:探讨秋水仙碱磁微球靶向治疗乳腺癌的可行性。方法:用白蛋白包埋法制备秋水仙碱磁微球,测量微球大小、载药量,进行药物的急性毒性实验和亚急性毒性实验,在外加磁场作用下观察其对裸鼠移植瘤抑制作用。结果:制得微球直径在200 nm左右,药物包裹率在37%以上,对乳腺癌EMT6细胞裸鼠移植瘤有显著抑制作用(抑瘤率30.2%),较游离秋水仙碱治疗组强(抑瘤率为27.6%)。结论:秋水仙碱磁微球外加磁场引导定位对乳腺癌有较好的治疗作用。 Objective: To probe into possibility in orientation of colchieine magnetic microspheres in treating breast cancer. Methods: Ferrimagnetic microspheres of human serum albumin colchicine(FM - HSA- Co) were prepared by human serum albumin containing methods. The size and drugs carried by microspherea were measured, acute toxic teat, subaeute toxic teat of drugs were performed. Inhibitory effect was observed on breast cancer EMT6 cell xenografts in nude mice with external magnetic orientation. Results: FM - HSA - Co's diameter was about 200nm, the drug contained ratio was over 37%. FM - HSA- Co and free colchieine in vivo the tumorinhibiting rate against breast cancer EMT_6 cellxenografts in nude mice were reapeetively 30.2% and 27.6% (P<0.05 vs control). Conclusion: With the assistance of external magnetic orientation, FM - HSA - Co has a stronger inhibition to breast tumor.
