摘要
目的:通过检测阳和汤对骨关节炎(OA)软骨细胞缺氧诱导因子1αmRNA(HIF1αmRNA)表达的影响,探讨阳和汤改善OA软骨退行性变的机制。方法:30只新西兰大白兔随机分为正常组、模型组、阳和汤组,每组10只,按照Hulth法建立的膝骨关节炎模型,分别对关节软骨病理切片后采用Safranin O染色,光镜下观察,进行Mankin评分,同时实时荧光定量PCR检测HIF1αmRNA在各组的表达。结果:正常对照组Safranin O染色的Mankin评分与模型组差异有统计学意义(P=0.005),阳和汤组Safranin O染色减少或缺失程度较模型组轻,差异有统计学意义(P=0.003)。模型组HIF1αmRNA表达较正常组明显高,差异有统计学意义(P=0.035),阳和汤组HIF1αmRNA表达明显低于模型组,差异有统计学意义(P=0.039)。结论:阳和汤可延缓关节软骨退行性变,其作用可能是通过调控HIF1αmRNA的作用。
Objective: To explore the influence of Yanghe decoction ( 阳和汤) on the expression of HIF- 1α mRNA in osteoarthritis (OA) ,so as to study mechanisms of Yanghe decoction ( 阳和汤 )in the improvement of cartilage degeneration. Methods: Thirty New Zealand white rabbits were randomly divided into 3 groups : control group ( 10 rabbits), OA model group ( 10 rabbits ) and Yanghe decoction ( 阳和汤)treatment group ( 10 rabbits ). The OA models were established by using Hulth method. The sections were stained with Safranin O for histological examination. Each sample was evaluated the cartilages histological characteristics according to the method of Mankin. Fluorescent quantitative PCR was used to detect the expression of HIF-1α mRNA in each group. Results:There were significant differences in the Mankin score between the control group and the OA model group (P=0.005) ,either between the OA model group and the Yanghe decoction ( 阳和汤)treatment group (P= 0.003 ). Fluorescent quantitative PCR indicated that HIF-1α mRNA significantly increased in the OA model group compared to that of the control group (P=-0.035), but that expression was much lower in Yanghe decoction ( 阳和汤 )treatment group compared to that of the OA model group (P=0.039). Conclusion:The expression of HIF-1α mRNA plays an important role in OA. Yanghe decoction (阳和汤)is effective to protect the articular cartilage, which is achieved through regulating the expression of HIF-1α mRNA.
