摘要
目的建立D-半乳糖性白内障大鼠动物模型,检测二肽基肽酶Ⅲ(DPPⅢ)在D-半乳糖性白内障大鼠晶状体和正常大鼠晶状体中的含量,探讨DPPⅢ与白内障发病的关系。方法建立D-半乳糖性白内障大鼠动物模型,进行相应分期分组。应用Westernblot方法检测DPPⅢ在各组白内障大鼠及正常大鼠晶状体中的含量。结果DPPⅢ在各组D-半乳糖性白内障大鼠晶状体中的含量明显高于同期正常对照组,并随着晶状体混浊程度的加重而增加。结论DPPⅢ在D-半乳糖性白内障大鼠晶状体中的含量较正常对照组增加,DPPⅢ含量的变化可能与白内障发病程度相关。
Objective Dipeptidyl peptidase Ⅲ( DPPⅢ ) exist widely in eytoplasm of muhiple tissues. It is involved in the metabolism of protein. This study was to establish the D-galactose cataraetous rat model and explore DPPⅢ content in cataractous lens and its significance. Methods The cataractous model was induced by intraperitonely injection of 50% D-galactose 30 g/(kg · d) two limes per dav in 40 Wistar rats. The animal models were assigned to the experimental group 1 and 2 respeclively according to the different degree of cataract following the modeling, and 20 normal rats were divided into 2 control groups. Western blot assay was used to detect the content of DPP Ⅲ in lens in the control group and model group. Results The lens of the control rats were transparent during the observation period,and the model rats showed lens opacification. Western blot analysis showed the stronger bands in the experimental rats compared with control rats. The total gray value of DPP Ⅲ in experimental rats was higher than that of the control rats,and that in experimenlal group 2 was higher than the experimental group 1. ConclusionThe protein level of DPP Ⅲ in the lens of cataractous rats is higher than that of corresponding normal control groups. The changes of protein level of DPPⅢ may play a role during cataraet formalion.
