摘要
目的:探讨比较沙棘总黄酮(TFH)和广枣总黄酮(TFC)对缺血心肌的保护作用及其相关蛋白表达谱的改变。方法:应用表面增强激光解吸离子化蛋白质芯片技术检测了经TFH和TFC处理后的缺血心肌组织和未经处理的的对照组缺血心肌组织中的蛋白质谱。用PHSII-C型蛋白芯片阅读机读取数据并进行比较分析。结果:与对照组的缺血心肌组织相比,经TFH处理后的缺血心肌组织3种芯片共捕获6个存在有差异的蛋白质峰,其中有1个呈高表达,有5个呈低表达;经TFC处理后的缺血心肌组织3种芯片共捕获7个存在有差异的蛋白质峰,其中有2个呈高表达,有5个呈低表达;TFH和TFC比较,各蛋白质表达峰的强度有所差别。结论:TFH和TFC均可从蛋白质水平进行调节从而产生抗心肌缺血作用,但它们所影响的蛋白种类、数量及作用机制均有所差别。
Object:Total Flavones of Hippophae Rhamnoides L. (TFH)and Total Flavones of Choerospondias Axillaris Fructus(TFC)had showed a protection on myocardial ischemic injuries. However, the molecular basis of such protection remains unclear. Comparing the action of the microarray analysis for the protein expression in myocardium provide a strong tool to explore the key protein candidates involved in the pathogenesis of ischemic injury. Methods:Surface enhanced laser desorption/ionization (SELDI) mass spectrometry with protein chip IMAC3, SAX2 and NP20 was used to compare the differentially expressed protein in TFH-treated, TFC-treated and untreated ischemic myocardium in rats and the results were analysized with Proteinchip Software 3.0.2. Results:Six differentially expressed proteins were identified in TFH treated myocardium and these differential effects correlated with the expression of five downregulated proteins and one upregulated proteins. Seven differentially expressed proteins were identified in TFC treated myocardium and these differential effects correlated with the expression of five downregulated proteins and two upregulated proteins. Conclusion: The myocardial protection of TFH and TFC may be mediated by the differencial expression of these proteins which could be the key protein candidates for different kinds and amounts.
