TR6mRNA在肝癌中的表达及其临床意义

EXPRESSION OF TR6 MRNA AND ITS CLINICAL SIGNIFICANCE IN HEPATOCELLULAR CARCINOMA

摘要 [目的]研究肝癌中TR6mRNA的表达及其与临床病理特征之间的关系,阐明TR6在肝癌转移过程中的作用。[方法]应用半定量RT-PCR方法,检测TR6mRNA在肝癌组织中的表达和相对含量,分析TR6mRNA的相对含量与肝癌临床病理特征的关系。[结果]69例肝癌组织中,67例标本可扩增出426bp的条带,TR6mRNA的阳性表达率为97.10%,癌旁组织TR6mRNA的阳性表达率为76.81%(53/69);癌组织中TR6mRNA相对含量明显高于癌旁组织(P﹤0.01)。TR6mRNA表达与转移复发和肿瘤大小有关,与年龄、性别、分化程度、临床分期、有无肝硬化、AFP、门脉癌栓、包膜浸润、肿瘤结节及HBsAg无关。序列分析发现其两例病例序列mRNA与源序列比较均有4个不同的碱基改变。[结论]肝癌中,TR6mRNA表达增高。肝癌TR6mRNA表达产物存在碱基改变。TR6mRNA的高表达在肝癌的转移复发过程中可能发挥重要的作用。 [Objective] To investigate the expression of TR6 mRNA and its correlation with clinicopathologic features of hepatocellular carcinoma(HCC)and illuminate its role in tumor metastasis.Methods The expression of TR6 mRNA and its relative amount were detected by semi-quantitive RT-PCR.The relationship between the relative amount of TR6 mRNA and clinicopathological features of HCC was analyzed.Results Among 69 cases of HCC tissues,a 426 bp band was detected in 67 cases,and the positive rate of TR6 mRNA expression was 97.10%.The rate of TR6 mRNA expression in adjacent-tumor liver tissues was 76.81%(53/69).The relative amount of TR6 mRNA in HCC was significantly higher than that in the adjacent-tumor liver tissues(P﹤0.01).The expression of TR6 mRNA was associated with metastasis and the tumor size but had no correlation with age,gender,differentiation grades,clinical stages,cirrhosis,AFP,portal vein tumor embolus,capsular infiltration,tumor nodes and HBsAg.Sequencing analysis showed 4 different point mutations in the DcR3 gene mRNA of HCC in two cases of positive amplification products.Conclusion TR6 mRNA is over-expressed in patients with HCC.Point mutation of the expression product of TR6 gene in mRNA exists in HCC.The over-expression of TR6 mRNA may play a vital role in the process of metastasis in HCC.
