摘要
目的探讨凋亡相关基因bcl-2和bax在羟基磷灰石(HAP)纳米粒子诱导肝癌SMMC-7721细胞凋亡后表达的变化。方法采用免疫组化法检测bcl-2蛋白和bax蛋白表达。结果不同浓度的HAP纳米粒子处理细胞24、48、72h后bcl-2蛋白的表达降低,bax蛋白的表达升高,与对照组比较均有显著性差异,并且均呈现时间和剂量依赖性。结论HAP纳米粒子诱导肝癌SMMC-7721细胞凋亡,与凋亡调控基因bax表达增强、bcl-2表达减少有关。
Objective To investigate the expression of bcl-2 and bax after liver cancer cell line SMMC-7721 apoptosis induced by hydroxyapatite nanoparticles. Methods The expressions of bcl-2 and bax protein were detected by immunohistochemical method. Results The expression of bcl-2 protein decreased, that of bax protein increased 24, 48 and 72 h after treated by different concentrations of hydroxyapatite nanoparticles, having significant difference from those of control, presenting time and dosage dependency. Conclusions Hydroxyapatite nanoparticles could induce liver cancer cell line SMMC-7721 apoptosis, which is related to bax expression increasing and bcl- 2 expression decreasing. :
