缬沙坦对糖尿病大鼠脑组织氧化应激及内皮功能的影响

摘要 目的探讨缬沙坦对糖尿病大鼠脑组织氧化应激及内皮功能的影响。方法用链脲佐菌素诱发糖尿病大鼠模型。24只大鼠随机分为正常对照组、糖尿病组、缬沙坦组(缬沙坦40mg·kg-1·d-1)。12w后测定各组体重、血糖,比色法测定脑组织中一氧化氮(NO)、丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,荧光实时定量RT-PCR检测内皮素(ET-1)mRNA、核因子-κB(NF-κB)mRNA表达。结果(1)第12周糖尿病组和缬沙坦组体重均明显轻于正常对照组(P<0·01),血糖在糖尿病组、缬沙坦组明显高于正常对照组(P<0·01),糖尿病组与缬沙坦组间无显著差异。(2)糖尿病组MDA水平显著高于缬沙坦组和正常对照组(P<0·01),而NO含量、SOD及GSH-Px活性显著低于缬沙坦组和正常对照组(P<0·01)。(3)糖尿病组ET-1 mRNA和NF-κB mRNA的表达量较正常对照组和缬沙坦组明显增多,缬沙坦组较正常对照组明显增多,较糖尿病组明显减少,各组间均有显著差异(P<0·01)。结论高糖内环境能促进糖尿病大鼠脑组织氧化应激水平,损害内皮功能,而缬沙坦能通过抑制NF-κB活化,增强抗氧化能力,逆转内皮功能损害,有利于改善脑功能。
