摘要
目的:通过检测大鼠眼挫伤前后视觉电生理的变化,探讨重组人促红细胞生成素(recombinant human erythropoietin,rhEPO)对挫伤眼视功能的保护作用。方法:将SD大鼠12只随机分为对照组及EPO组,均选取左眼为实验眼。造模前先进行左眼闪光视网膜电图(flash electroretinogram,FERG)和闪光视觉诱发电位(flash visual evoked potentials,FVEP)检查,之后采用重击法致2mo龄SD雄性大鼠(体质量180~220g)左眼挫伤,伤后立即给予EPO组注射rhEPO:5U(5μL),对照组玻璃体注射同等剂量生理盐水,伤后1,4,7,14d分别进行左眼FERG和FVEP检查。结果:造模前两组的FERG-b波和FVEP-P100波的振幅值无显著性差异(P〉0.05)。重击法造模后,对照组实验眼的FERG-b波和FVEP-P100波的振幅值在伤后第1d均降至最低水平,以后逐渐恢复,第14d未能恢复造模前水平(P〈0.05)。但EPO组与对照组FERG-b波和FVEP-P100波的振幅值差异明显,实验结束时EPO组左眼FERG-b波和FVEP-P100波的振幅值明显高于对照组(P〈0.01),接近造模前水平(P〉0.05)。结论:rhEPO可以显著改善大鼠眼挫伤后视网膜缺血时的FERG-b波和FVEP-P100波振幅,起到保护视功能的作用。
AIM :Through detecting the electrophysiology, including the changes in flash electroretinogram (FERG) and flash visual evoked potentials (FVEP), we investigated the protective effect of recombinant human erythropoietin (rhEPO) in the rat retina after the eye contusion made by modified Allen's failing strike.
METHODS:Twelve SD rats were randomly divided into the control group(n=6) and the EPO group (n=6). All the left eyes of the control group and the EPO group were made into the contusion model by modified Allen's failing strike. After the model was made, rhEPO 5U (5μL) was injected into the vitreous of EPO group rats. The isodose of normal saline was injected into the vitreous of the control group rats. FERG and FVEP were detected at 30 minutes, before and on the 1st,4th,Tth and 14th day after contusion respectively.
RESULTS: The amplitude of FERG-b wave and the amplitude of FVEP-P100 wave had no significant difference between the control group and the EPO group before ocular contusion ( P 〉 0.05). The amplitude of FERG-b wave and the amplitude of FVEP-Ple0 wave of the control group dropped down to the lowest level on day 1 after contusion and continued to recover thereafter, However, they can' t come back to the level before being struck till day 14( P〈0.05).The resembling situation was in the EPO group except that the amplitude of FERG-b and FVEP-P100 wave came back to the level before being struck at day 14 ( P 〉 0.05). But there were obvious differences between the two groups at the amplitude of FERG-b and FVEP-P100 wave. The EPO group's is higher than the control group's (P〈0.01).
CONCLUSION. rhEPO can improve the amplitude of FERG-b wave and FVEP-P100 wave following ocular contusion. This indicates that rhEPO has the potential to be an optimal neuroprotective agent.
