摘要
目的研究大孔树脂吸附法富集一枝蒿总黄酮的工艺条件及参数。方法以总黄酮为考察指标,用比色法测定含量,考察大孔吸附树脂富集一枝蒿总黄酮的工艺条件。结果一枝蒿提取液(0.04g生药/ml)10ml上大孔树脂柱(R15mm×H290mm),吸附30min后,先用150ml蒸馏水洗脱,然后,采用150ml 70%乙醇洗脱,洗脱速度为3ml/min,洗脱剂用量为10倍量树脂,树脂可重复使用3次,采用此条件为最佳工艺。结论HPD-450型大孔树脂在所确定的工艺条件下,可较好地吸附分离一枝蒿总黄酮。其洗脱物中总黄酮含量达3.18%以上,总黄酮回收率为86.91%以上。
Objective To study the optimal conditions and the technological parameters for enrichment process of total flavone of Artemisia rupestris L. with maeroporous absorption resin. Methods With the enrichment degree of total flavone of Artemisia rupestris s L.. as a marker, the optimal conditions and technological parameters of the enrichment process were investigated. Results The optimum process was as follows : the concentration of extract of water from the Artemisia rupestris L. was 0.04 g/ml; eluting velocity, 3 mE/ min; the eluting reagent, 70% ethanol ( 10 times the volume of the resin) ; and the HPD -450 resin could be used 3 times repeatedly. Conclusion The results showed that HPD - 450 macroporous resin could well absorb and separate Artemisia rupestris L. total flavone under the set technological conditions, and the content of total flavone in Artemisia rupestris L. is over 3.18% , with a total flavone recovery rate of over 86.91%.
