促红细胞生成素对颅脑创伤休克大鼠的保护作用 被引量:5

Protective role of Erythropoietin for traumatic brain injury with shock in rats

摘要 目的探讨重组人促红细胞生成素(r-HuEPO)对大鼠伴有休克的颅脑创伤的保护作用。方法114只Wistar大鼠随机分为假手术组(n=6)、单纯脑创伤组(n=36)、合并休克的脑创伤组(n=36)及r-HuEPO治疗组(n=36)。采用自由落体法建立大鼠颅脑创伤模型,股动脉放血造成休克模型。分别于伤后不同时间点,应用Epics XL流式细胞仪检测颅脑创伤后神经细胞凋亡情况,利用Caspase-3分光光度法检测试剂盒测定Caspase-3活性变化,并用干湿重法测定脑组织含水量。结果单纯脑创伤组及合并休克的脑创伤组细胞凋亡率、Caspase-3活性及脑组织含水量均较假手术组明显升高(P〈0.05)。与模型组相比,r-HuEPO治疗组Caspase-3活性及细胞凋亡率显著降低(P〈0.05),脑水肿虽有好转,但无统计学意义(P〉0.05)。结论r-HuEPO可通过降低Caspase-3的活性,减少细胞凋亡,起到脑保护作用。 Objective To investigate the effects of r-HuEPO on brain prevention after traumatic brain injury,(TBI) with shock in rats. Method One hundred and fourteen adult Wistar rats were randomly divided into sham group( n = 6 ), TBI group(n=36), TBI with shock group(n=36) and r-HuEPO group( n = 36). The experimental TBI model was established by Feeney's method, and the shock model was established by exsanguinating femoral artery. Samples were harvested after injury for measuring apoptosis of cells by Epics XL Flow Cytometer, determining the activity of Caspase-3 by Caspase-3 Colorimetric Assay Kit and measuring the water content of injuried brain by dry-wet weight method. Results Caspase-3 activation, the number of apoptotic cell and the water content of injuried brain were all increased significantly in TBI group and TBI with shock group (P〈0.05). r-HuEPO significantly down-regulation Caspase-3 activation and decreased the apoptotic rates(P〈0.05). There was a slight but not significantly better brain edema in treatment group compared to TBI with shock group(P〉0.05). Conclusions r-HuEPO might be useful to protect brain against damage by inhibiting caspase-3 activation and apoptosis.
