摘要
目的:研究红芪总多糖(THPS)诱导小鼠S180瘤细胞凋亡作用及其机制。方法:昆明小鼠60只接种S180瘤后随机分为5组。(1)模型组(S):生理盐水腹腔注射,(2)环磷酰胺组(CY):CY腹腔注射,(3)THPS低剂量(H100),(4)THPS中剂量(H200)和(5)THPS高剂量组(H400):分别给予THPS低剂量、中剂量和高剂量腹腔注射,14 d后处死小鼠。电镜观察各组瘤细胞的形态,激光共聚焦(LSCM)测定每组瘤细胞DNA、RNA含量、细胞内钙离子浓度、自由基含量及线粒体膜电位。结果:中剂量的THPS可明显升高细胞内钙离子浓度、自由基含量及降低线粒体膜电位和瘤细胞DNARNA含量的作用。结论:THPS中剂量通过升高细胞内钙离子浓度、自由基含量及降低线粒体膜电位诱导小鼠S180瘤细胞凋亡。
AIM: To study effects and mechanisms of total hedysari polysaccharide (THPS)inducing apoptosis of S180 tumor. METHODS : 60 Kunming mice were divided into 5 groups randomly after inoculating S180 tumor. Sample group (S) and cyclophosphamide group (CY) were injected intraperitoneally with normal sodium and cyclophosphamide respectively. Low dose THPS group (H100), moderate dose THPS group (H200), and high dose THPS group (H400) were injected intraperitoneally with low, moderate, and high dose THPS respectively. The mice were killed after being treated for 14 days. Appearance of tumor was observed by electron microscope, and DNA, RNA, intracellular Ca^2+ , free radicals, mitochondrial membrane potential of tumor were determined by laser scanning confocal microscope. RESULTS : Moderate dose THPS raised intracellular Ca^2+ , free radicals and reduced mitochondrial membrane potential and DNA, RNA of tumor. CONCLUSION: Moderate dose THPS induces apoptosis of S180 tumor through raising intracellular Ca^2+ , free radicals and reducing mitochondrial membrane potential.
