摘要
采用正交试验法研究黑果枸杞类黄酮提取和精制最佳工艺条件。结果表明黑果枸杞叶中类黄酮含量远远大于枝中,从叶中提取最佳工艺条件为:以60%乙醇作提取剂、固液比1:20、60℃提取40 min,类黄酮提取率为0.228 9%,类黄酮含量为0.415 6%;枝中提取最佳条件为:以80%乙醇作提取剂、固液比1:30、70℃提取80 min,类黄酮提取率为0.100 4%,类黄酮含量为0.303 5%;采用AB-8型大孔树脂对叶类黄酮进行精制,精制条件为:上样液浓度0.200 g/L、流速2 mL/min、pH=5.0,以95%乙醇洗脱、流速4 mL/min、用量为5倍柱床体积。经树脂纯化后的精制品中类黄酮含量达到28.0%,含量比粗制品提高了67.37倍。说明该工艺可行,可应用于实际生产。
The extracting and refining technology of total flavonoids of Lycium ruthenicum Murr.was studied by orthogonal design in this paper. Experimental results showed that flavonoids content of leaves was more than of branches, the best extracting conditions were 60 % ethanol at 60 ℃ for 40 min with the solution and solid ratio was 1:20 to leaves. Extraction ratio of flavonoids in leaves was 0.228 9 %,flavonoids content of crude product of leaves was 0.415 6 %.While the best extracting conditions to branches were 80 % ethanol at 70 ℃ for 80 minwith the solution and solid ratio was 1:30. Extraction ratio of flavonoids in branches was 0.100 4 %,flavonoids content of crude product of branches was 0.303 5 %. Refining crude flavonoids of leaves used AB-8 resin, the optimum refining conditions were: the concentration of sample solution was 0.200 g/L,velocity of sample solution was 2 mL/min,pH value was 5.0, eluting with 95 % ethanol from AB-8 resin,velocity of ethanol was 4 mL/min, volume of ethanol was 5 times volume of resin.The purity of product reached 28.0 % and improved 67.37 times than crude product,these methods were sutable and applicable to practice manufacturing.
