人食管鳞状细胞癌组织中KiSS-1 mRNA及蛋白的表达 被引量:5

摘要 目的:探讨食管鳞状细胞癌(ESCC)组织中KiSS-1mRNA及蛋白的表达情况。方法:分别采用原位杂交方法和免疫组织化学SP法对49例ESCC、26例癌旁不典型增生组织及49例正常食管黏膜组织进行KiSS-1mRNA和蛋白的检测。结果:ESCC组织、癌旁不典型增生组织及正常食管黏膜组织中KiSS-1mRNA的阳性表达率分别为61.2%(30/49)、80.8%(21/26)、95.6%(47/49),依次升高(χ2=17.856,P<0.01);Kiss-1蛋白的阳性表达率分别为69.4%(34/49)、80.8%(21/26)、98.0%(48/49),依次增高(χ2=14.340,P<0.01);ESCC组织中KiSS-1mRNA及蛋白表达与食管癌组织学分级、浸润深度无关(P>0.05),与淋巴结转移有关(P<0.05);ESCC组织中KiSS-1mRNA与蛋白的表达呈正相关(rs=0.653,P<0.05)。结论:KiSS-1表达缺失与ESCC的发生、转移有关;KiSS-1有望成为ESCC早期诊断和判断预后的辅助指标。 Aim : To investigate the expression of KISS-1 mRNA and protein in esophageal squamous carcinoma ( ES- CC) tissue. Methods:The expression of KISS-1 mRNA and protein in 49 cases of ESCC, 26 cases of adjacent atypical hy- perplasia epithelium and 49 cases of normal esophageal epithelium tissues were detected by in situ hybridization and SP im- munohistochemical method,respectively. Results: The expression rates of KISS-1 mRNA in ESCC, adjacent atypical hyper- plasia epithelium and normal esophageal epithelium tissue, were 61.2 % ( 30/49 ) ,80.8 % ( 21/26 ) ,95.6% ( 47/49 ) , re- spectively,and increased by turns(x2 = 17. 856,P 〈 0.01 ). The expression rates of KiSS-I protein in carcinoma, adjacent atypical hyperplasia epithelium and normal esophageal epithelium tissue, were 69.4% (34/49), 80.8% (21/26) ,98.0% (48/49) , respectively,and increased by turns(x2 = 14. 340 ,P 〈 0.01 ). The mRNA and protein expression of KISS-1 in ESCC tissue were correlated with lymphatic metastasis (P 〈.0.05 ), hut not with infilatration and tumor grade( P 〉 0.05 ). There was positive correlation between the protein and mRNA expression of KISS-1 in ESCC tissue ( r = 0. 653 ,P 〈 0.05 ). Conclusion: The loss expression of KISS-1 is closely correlated to the formation and metastasis of ESCC, and KISS-1 may be a new assistant index for early diagnosis and prognosis for ESCC.
