Effects of nerve growth factor on the regeneration of neurocytes after the hypoxia-ischemic brain damage of neonatal rats

摘要 目的探讨外源性神经生长因子(NGF)对新生鼠缺氧缺血性脑损伤(HIBD)后神经再生的影响。方法通过7日龄新生Wistar大鼠制作HIBD模型,设置假手术组、HIBD对照组、NGF治疗组,应用5-溴脱氧尿嘧啶(BrdU)标记新生神经细胞,免疫组化方法和计算机图像分析技术检测不同时点(术后4、7、14、21d)海马齿状回(DG)巢蛋白(nestin)表达、BrdU阳性细胞数的变化。结果各时点HIBD对照组nestin、BrdU阳性细胞数较假手术组增加(P均<0.05);各时点NGF治疗组nestin、BrdU阳性细胞数较假手术组和HIBD对照组增加(P均<0.01)。结论早期给予外源性NGF治疗可促使新生鼠脑缺氧缺血后中枢神经系统nestin、BrdU的表达增加,NGF治疗可促进受损神经细胞的增殖再生,在神经细胞损伤的修复中发挥一定的保护作用。 Objectives To investigate the effects of nerve growth factor (NGF) on the regeneration of neurocytes following hypoxia-ischemic brain damage (HIBD) of neonatal rats. Methods The HIBD model was set up in seven-day-old newborn Wistar rats, and the rats were divided into two groups randomly, the NGF treatment group and the control group. A sham operation group is included. The newly generated neurocytes were labeled by bromodeoxyuridine (BrdU). The expressions of nestin in hippocampal dentate gyrus (DG) and the number of BrdU positive cells were examined by immunohistochemical staining and image quantitative analysis respectively at 4 th, 7 th, 14 th, 21 th days after the operation. Results The number of nestin and BrdU positive cells was significantly increased in HIBD rats than in sham operation group at all time points (P 〈 0.05), and was also significantly increased in NGF treatment group than control group of HIBD at all time points (P 〈 0.01 ). Conclusions Exogenous NGF could enhance the expression of nestin and BrdU in hippocampal DG region of neonatal rats with HIBD, and may play an important role in the regeneration of neurocytes damaged by hypoxia-ischemia.
