摘要
目的探讨C225单用及与顺铂(DDP)联用对于人子宫颈癌HeLa细胞裸鼠移植瘤生长的抑制作用及机制。方法将40只子宫颈癌HeLa细胞株裸鼠移植瘤模型随机分为实验对照组、C225组、DDP组和C225+DDP组,每组10只。各组裸鼠分别经腹腔注射生理盐水、C225(1mg/只·次)、DDP(5mg/kg·次)、和C225+DDP(C2251mg/只·次,DDP5mg/kg·次)每周2次,共4周。治疗期间定期测量肿瘤大小,观察裸鼠全身状况,实验结束时,将移植瘤完整取出,称取瘤重,计算抑瘤率,并用免疫组织化学技术检测各组肿瘤组织中PCNA、VEGF的表达。结果C225组、DDP组、C225+DDP组肿瘤的平均体积均显著小于实验对照组(P=0.008,P=0.001,P=0.000);C225+DDP组的肿瘤体积亦显著小于DDP组与C225组(P=0.025,P=0.014);各组平均瘤重分别为2.67±0.18g、1.84±0.37g、1.57±0.18g、1.2±0.09g,C225组、DDP组、C225+DDP组与实验对照组之间具有显著性差异(P=0.015,P=0.001,P=0.000),抑瘤率分别为31.09%、41.2%、55.06%。免疫组化结果显示,PCNA阳性表达率在C225组、DDP组与C225+DDP组较实验对照组显著降低(P=0.033,P=0.011,P=0.003);VEGF的表达率经C225与DDP单用及联合治疗后较实验对照组显著降低(P=0.033,P=0.033,P=0.003)。结论C225与DDP均可抑制人子宫颈癌HeLa细胞株裸鼠移植瘤生长,两药联合应用的抑瘤效果优于两药单用;C225单用及与DDP联用后可显著抑制PCNA与VEGF的表达水平。
Objective To explore the inhibitive effect on the growth of transplanted tumor of HeLa cells in nude mice after treatment of C225 and iC225 combined DDP. Methods Forty nude mice with transplanted tumor were divided into four groups in radam: normal control group, C225 group, the DDP group and C225 + DDP groups, each group had 10 mice. All groups of nude mice were respectively injected with normal saline(NS), C225 (1 mg each time), DDP (5 mg/kg each time) and C225 + DDP (C225 1 mg and DDP 5 mg/kg each time), twice every week, for four weeks. During the period of treatment, the size and volume of tumor were measured and the systemic conditions of the mice were observed. At the end of the trial, the tumor was removed and the volume and weight of tumor were mesured. The inhibitive rate was caculated and the expression of PCNA and VEGF was detected with immunohistochemical technology. Results The tumors of all treatment groups were smaller than the control group(P = 0. 008,0. 001 and 0. 000, respectively). The volume of the tumors of the combined group were smaller than the DDP group and C225 group(P = 0. 025, P = 0. 014). The average weight of tumor was 2.67 ± 0. 18 g,1.84 ± 0. 37 g,1.57 ± 0.18 g and 1.2 ± 0. 09 g, respectively. The weight of C225 group,DDP group and C225 + DDP group were lighter than that of the control group(P = 0. 015, 0. 001 and 0. 000, respectively) ;The inhibitive rates of C225 group, DDP group and C225 + DDP group were 31.09% ,41.2% and 55.06%. The results of immunohistochemical technology showed that the expression rate of PCNA in C225 group, DDP group and C225 + DDP group decreased significantly compared to the control group(P〈0.05, P〈0.01, P〈0.01 ). The expression of VEGF in C225 group, DDP group and C225 + DDP group decreased compared to that of the control group(P〈0.05 ,P〈0.01 and P〈0.01). Conclusions C225 and DDP can inhibit the growth of transplanted tumor of HeLa cell lines in nude mice. The effect of C225 + DDP is better than that of C225 and DDP. The treatment of C225 and C225 combined DDP can significantly inhibit the expression of PCNA and VEGF.
