朝鲜族人群DXS6854基因座的遗传多态性研究

摘要 [目的]探讨吉林省延边地区朝鲜族人群短串联重复序列(STR)基因座DXS 6854的遗传多态性分布,获取相应多态位点的群体遗传学数据.[方法]采用PCR扩增技术及聚丙烯酰胺凝胶电泳方法,分析101名朝鲜族女性和160名朝鲜族男性个体的DXS 6854 STR基因座等位基因频率.[结果]在女性样本DXS 6854基因座中各检测出6种等位基因和17种不同的基因型,基因座多态性分布符合Hardy-Weinberg平衡定律,杂合度为0.772 3,多态信息含量为0.67,个人识别率为0.857;在男性样本DXS 6854基因座中检测出6种等位基因,单倍型多样性为0.734.[结论]DXS 6854基因座具有较高的个人识别力、杂合度及多态信息量,是较理想的遗传标记系统,对朝鲜族群体的个人识别、亲子鉴定及民族遗传学研究具有重要意义. OBJECTIVETo understand the genetic polymorphism at DXS6854 short tandem repeat(STR) locus in the Korean-Chinese of Yanbian area,and to construct a corresponding database of population genetics.METHODSThe allele frequencies of the DXS6854 locus in 101 of female and 160 of male with no sibship,in the Korean-Chinese in Yanbian area were analyzed by PCR and polyacrylamide gel electrophoresis(PAGE).RESULTSSix alleles and seventeen genotypes were observed at DXS6854 locus in the female individuals,respectively,and met Hardy-Weinberg equilibrium.The statistical analysis of DXS6854 STR locus showed the heterozygosity was 0.7723,the polymorphic information contents(PIC) was 0.67;the power of discrimination(PD) was 0.857;and also six alleles were observed at DXS6854 locus of the male individuals,the haplotype diversity reached 0.734.CONCLUSIONDXS6854 STR locus has relative high heterozygosity and PIC,and so it could provide useful markers for genetic purposes,can be used in forensic identification and paternity.
