摘要
目的:观察阳离子脂质体介导反义血管内皮生长因子(VEGF)寡核苷酸对肺癌小鼠移植瘤生长、转移的抑制作用。方法:取肺癌细胞悬液建立小鼠肺癌动物模型后,选择40只随机分成4组,分别为空白对照组、实验对照组、VEGF反义寡核苷酸(ASODN)治疗组和VEGF正义寡核苷酸(SODN)治疗组。治疗结束后行大体、HE染色观察,并观察小鼠营养状况和症状。结果:对照组、VEGF反义寡核苷酸治疗组和VEGF正义寡核苷酸治疗组平均瘤质量分别为(7.14±1.68)、(4.26±1.37)和(7.33±1.56)g,VEGF反义寡核苷酸治疗组、VEGF正义寡核苷酸治疗组抑瘤率分别为58.2%和7.4%,ASODN组与对照组及SODN组比较差异有统计学意义,χ2分别为8.21和8.04,P值均<0.01。结论:阳离子脂质体介导反义VEGF寡核苷酸对小鼠肺癌的生长具有显著抑制作用,未发现明显的毒副反应,阳离子脂质体介导的基因转染技术有望成为治疗癌症的新方法。
OBJECTIVE:To evaluate the inhibitory effection of antisense vascular endothelial growth factor oligonucleotides on lewis lung cancer in a mice model by sa cationic liposomes. METHODS:The 40 nice of the established Lewis lung cancer model were divided into four groups randomly: VEGF-ASODN group, VEGF-SODN group and two control groups. The weight and volume of subcutaneous tumors were measured,and the morphological changes of tumor cells were observed under a microscope. RESULTS: The mean tumor weight of the VEGF-ASODN group was (4.26±1.37) g, that of the VEGF-SODN group was (7.33±1.56) g, and that of the control group was (7. 14± 1.68 ) g. The inhibition rates of tumor growth of the VEGF-ASODN group and VEGF-SODN group were 58.2% and 7.4%, respectively. Compared with the control groups, the VEGF-ASODN group showed significance in tumor size and inhibition rates· Х^2 were 8.21 and 8.04 ,P〈0.01. CONCLUSIONS: The Lewis lung cancer can be inhibited powerfully by antisense VEGF oligonudeotides combined with cationic liposomes. Little side effects are found. The technic of transfection by sa cationic liposomes might he a new method for therapy of cancer.
