养目平突汤含药血清对人眼球后成纤维细胞的影响 被引量:2

Effects of Yangmupingtutang Containing serum on cultured retroocular fibroblasts in patients with Graves ophthalmopathy

摘要 目的观察养目平突汤含药血清对人眼球后成纤维细胞(RFS)增殖以及合成透明质酸、Ⅰ型胶原蛋白的影响。方法体外培养正常人RFs,予养目平突汤含药血清孵育后,MTT比色法检测RFs的增殖;放免法检测透明质酸(HA)和Ⅰ型胶原蛋白的合成状况。结果正常组A值为(0.39±0.05)D,而养目平突汤含药血清各剂量组A值均减少,分别为(0.37±0.06)D(、0.30±0.05)D和(0.22±0.06)D(P均<0.05);正常组HA和Ⅰ型胶原蛋白的浓度分别为(1.64±0.12)mg/L、(1.72±0.11)mg/L;养目平突汤含药血清各剂量组HA和Ⅰ型胶原蛋白的合成均显著减少(P均<0.05),HA的浓度分别为(1.47±0.16)mg/L、(0.94±0.12)mg/L、(0.89±0.14)mg/L,Ⅰ型胶原蛋白的浓度分别为(1.55±0.12)mg/L(、0.98±0.13)mg/L和(0.90±0.10)mg/L。结论养目平突汤含药血清治疗Graves’眼病的作用途径部分是通过抑制RFs的增殖和HA、Ⅰ型胶原蛋白的合成,降低RFs生物活性,从而减少Graves’眼病的免疫损伤。 Objective It is to observe the influence of Yangmupingtutang Containing serum on cell proliferation, synthesis of hyaluronic acid (HA) and collagen I on cultured retroocular fibroblasts (RFs) in patients with Graves ophthalmopathy. Methods After cultured RFs were incubated with Yangmupingtutang Containing serum, RFs proliferatio was assessed by MTT, synthesis of HA and collagen I were measured by radioimmunoassay. Results The A value of nomal group was (0.39±0.05)D. The A values were decreased in the groups of Yangmupingtutang Containing serum, they were (0.37±0.06)D, (0.30±0.05 )D and (0.22±0.06)D respective. The synthesis of HA and Collagen I of nomal group were ( 1.64±0.12) mg/L, (1.72±0. 11)mg/L; and the synthesis of HA and Collagen I were significance decreased in the groups of Yangmupingtutang Containing serum (P〈0.05), the levels of HA were (1.55±0.12) mg/L, (0.98±0.13)mg/L and (0.90±0.10)mg/L respective, the levels of collagen I were (1.55±0.12)mg/L, (0.98±0.13)mg/L and (0.90±0.10mg/L. Conclusion Yangmupingtutang Containing serum can decrease the immune injury of Graves ophthalmopathy depend on inhibit the cell proliferation, the synthesis of HA and Collagen I , thereby decrease the bioactive of RFs.
