摘要
背景与目的:血管内皮生长因子(vascular endothelial growth factor,VEGF)在胶质瘤的血管生成和浸润性生长中发挥重大作用。本文研究人参皂苷Rg3对大鼠C6胶质瘤VEGF表达的影响,探讨其作为胶质瘤抗血管生成治疗的效果。方法:制备大鼠脑C6胶质瘤模型,在接种细胞模型建立2周后对治疗组给予人参皂苷Rg3治疗,同时设对照组和空白组,4周后处死动物观察肿瘤形成情况,Envision法进行VEGF免疫组织化学检测分析组间差异。结果:人参皂苷Rg3治疗组VEGF表达阳性和强阳性比例为15%,对照组阳性和强阳性比例为85%,空白组阳性和强阳性比例为85%;利用非参数Kruskal-Wallis检验进行统计学处理后发现人参皂苷Rg3治疗组VEGF表达较对照组明显减弱(P<0.01),人参皂苷Rg3治疗组VEGF表达较空白组明显减弱(P<0.01),对照组与空白组之间VEGF表达差异无统计学意义(P>0.05)。结论:人参皂苷Rg3可抑制脑胶质瘤VEGF表达具备抗血管生成作用。
BACKGROND & OBJECTIVE:To investigate the influence of Panaxsaponin Rg3 on VEGF expression in C6 glioma rat models, and explore its antiangiogenic effect. METHODS:C6 glioma rat models were established and devided into drug-treatment group, placebo-treatment group and blank group. Panaxsaponin Rg3 was administered 2 weeks after the intracranial injection of C6 cells in rats. Rats were sacrificed 4 weeks after Panaxsaponin Rg3 administration. Tumor size was measured and VEGF expression was examined using immunohistochemistry. RESULTS:The positive rate of VEGF in Panaxsaponin Rg3-treated group, placebo-treated group and blank group was 15%, 85% and 85% respectively. There was a significant statistical difference of VEGF expression level between Panaxsaponin Rg3-treated group and both placebo-treated group and blank group(P〈0.01). CONCLUSIONS :Panaxsaponin Rg3 can inhibit VEGF expression and angiogenesis in C6 glioma rat models.
