电针对老年性痴呆模型大鼠海马神经元凋亡蛋白Bcl-2和Cyt-c的影响 被引量:3

Effects of Electro-acupuncture Therapy on BcL-2 and Cyt-c in Hippocampus Neuron of Rats with Alzheimers' Disease

摘要 目的:研究电针对老年性痴呆(AD)模型大鼠海马区神经元中Bcl-2和Cyt-c的影响,从细胞凋亡的角度探讨电针治疗AD的部分作用机制。方法:以D-半乳糖腹腔注射和Aβ1-40海马注射诱导形成的AD模型大鼠为研究对象,电针"百会"、"涌泉"二穴,每日1次,连续20d。以通道式水迷宫测试学习记忆能力的变化,评价电针对AD的治疗效应;以免疫组化SP法检测海马神经元中Bcl-2和Ccy-c的表达。结果:与模型组大鼠相比,进入盲端的错误次数和平均逃避潜伏期明显减少(P<0.05),海马神经元中Bcl-2的阳性细胞数、积分光密度总和显著减少(P<0.05),而模针组大鼠海马神经元细胞色素C的阳性细胞数、积分光密度总和显著增加(P<0.05)。结论:电针能有效改善AD大鼠的学习记忆障碍,可能是通过促进AD大鼠海马神经元Bcl-2的表达,降低Cyt-c的表达,抑制细胞的凋亡,减少神经元的丢失来实现的。 Objective: To study the effects of electro-acupuncture therapy on BcL-2 and Cyt-c in hippocampus neuron of rats with Alzheimers' disease,and to probe into the mechanism of electro-acupuncture therapy in treating AD from the perspective of cell apoptosis.Methods: The AD model was established by abdominal injecting the D-galactose first and then hippocampus injecting β-amyloid 1-40.Then,Baihu Point(Du20) and Yongquan Point(KI1) were electro-acupunctured once a day for 20 days.The effects on the learning and memory ability of AD rats were observed by channel water maze,and then the expressions of protein BcL-2 and Cyt-c in hippocampus were detected by using immunohistochemistry method.Results: In comparison with control group,the times of rat's entering the blinded-end and average periods of swimming were decreased obviously than those in the treated group(P〈0.05).The expression of Bcl-2 in the hippocampus neuron of the rats in the treated group were decreased obviously(P〈0.05),and that of Cyt-c was enhanced(P〈0.05).Conclusion: Electro-acupuncture can improve the learning and memory abilities of AD rats,and the effects may be achieved by increasing the activities of BcL-2 and decreasing the expression of Cyt-c to inhibit the apoptosis,and then to prevent the neuron loss.
