摘要
目的探索一种正确的BMSCs抽取﹑分离和体外培养方法,建立BMSCs体外培养体系。方法倒置相差显微镜观察BMSCs细胞生长情况及形态变化特点。结果培养的BMSCs呈多种形态,有粗细不均的胞质突起,并且呈集落样生长。二代细胞形态更加单一,呈均匀生长,细胞因相互接触而产生接触抑制。大白鼠BMSCs经体外培养后生长活跃,具有良好的形态特征及生长特点。结论全骨髓贴壁培养法是分离BMSCs的一种比较理想的方法。
Objective To explore a kind of method of extraction, separation and cultivation of BMSCs in vitro, and establish a culture system of BMSCs. Methods The growth conditions and shape change characteristics of the BMSCs were observed by inverted phase contrast microscope in vitro. Results The cultivated BMSCs assumed many kinds of shapes, and showed the thick or thin uneven cytoplasmic process and the growth condition of colony type. The cellular shapes of the second generation were more unitary and assumed the even growth. The cells generated contact inhibition because of contact mutually. The BMSCs from SD rats through cultivation in vitro grow actively, had the good shape and growth characteristics. Conclusion The bone marrow adherence cultivation is a more successful method of separating BMSCs.
