高效液相色谱法测定谷物中的柄曲菌素 被引量:2

Determination of Sterigmatocystin in Cereals by Reversed-Phase High Performance Liquid Chromatography

摘要 建立了用反相高效液相色谱(RP~HPLC)法测定谷物中柄曲菌素(Sterigmato~cystin,ST)的含量。采用酸化乙腈震荡提取,C18固相萃取小柱净化,HPLC分离测定。样品中被测物与干扰物能得到良好的分离。柄曲菌素在10~1000μg/kg范围内与其峰面积呈良好的线性关系(r^2≥0.9999),回收率为88.4%~99.9%,相对标准偏差小于2.8%。检出限(S/N=3)为0.2ng,定量限(S/N=10)为0.5ng。该法简便、快速、准确,可用于谷物中柄曲菌素的检测。 A method was established for the determination of sterigmatocystin in cereals by reversed-phase high performance liquid chromatograph (RP-HPLC) after extraction by Ca8 cartridge. Separation and quantification were achieved by using a Agilent C18 column (250×4.6 mm i. d.,5 /lm) at 25℃, a mobile phase of acetonitrile and water(55:45) with a flow rate of 1 mL/min, and monitored with a diode-array detector at 325 nm. The linear range was from 10μg/L to 1 000μg/L with a detection limit of 10μg/L. The recoveries were 88.4%-99.9% with the relative standard deviations (RSDs) of 0.4%-2.8%. This method is simple, fast, accurate, convenient and could be used for quantitative determination of sterigmatocystin in cereals.
