摘要
应用兔大脑中动脉阻断模型,分离出突触膜(synapticplasmamembrane,SPM),线粒体(mitochondria,Mit)并测其Na+-K+-ATP酶活性,同时以放免法测定脑组织内皮素-1(ET-1)含量。结果发现脑缺血后脑组织ET-1含量显著增加,SPM、MitNa+-K+-ATP酶活性下降,两者呈显著负相关(r1=-0.9776,P<0.05;r2=-0.9980,P<0.02).提示ET-1在脑缺血脑水肿中的作用可能与其抑制Na+-K--ATP酶活性有关。
In the model of rabbit middle cerebral artery occlusion (MCAO), the synaptic plasma membrae (SPM), and mitochondria (Mit) were dissected and the activity of Na+ -K+ - ATPase in them and the contents of ET - 1 in brain tissue were determined. The content of ET - 1 was significantly increased and the activity of Na+-K+ - ATPase in SPM and Mit were decreased after cerebral ischemia, and there was negative correlation between content of ET- 1 and the activity of Na+-K+ - ATPase in SPM and Mit (r1= -0.9776, P<0.05;r,= -0.9980,P<0.02 respectively). These results suggested that the effect of ET - 1 on ischemic brain edema was associated with its inhibition of Na+ - K+ - ATPase activity in SPM and Mit.
