摘要
目的探讨光卟啉不同孵育浓度和不同孵育时间对人食管癌细胞Eca-109体外光动力治疗(pho-todynamic therapy,PDT)效应的影响。方法以不同浓度光卟啉孵育Eca-109细胞,并给予不同孵育时间(4,6,12,24h)(光源采用DIOMED630PDT系统)后于饱和光能量密度20J/cm2下行PDT,24h后通过MTT法检测细胞生存率。结果光卟啉4种不同孵育时间和不同孵育浓度间生存率差异均有显著性(均P<0.01)。同一孵育时间下不同浓度间的生存率差异均有显著性(均P<0.01),而同一浓度下不同孵育时间之间的生存率,则除了浓度为0μg/ml和0.1μg/ml外差异均有显著性(均P<0.05)。结论特定光源下,光卟啉的孵育浓度和孵育时间对人食管癌细胞Eca-109体外PDT效应影响显著。
Objective To investigate the influence of phortofrin photodynamic therapy effect with different incu- bation concentration and incubating time in human esophageal cancer cell line in vitro. Methods Human Eta - 109 cells were incubated in vitro with phortofrin in different incubating concentration, and followed by irradiation in 20 J/m2 saturate light energy density under DIOMED630 PDT systems. Survival rate of each concentration was measured by MTT assay after 24 hours incubation. Results The cell survival rate between different phortofrin incubating concentration and different incubating time and under the same incubating time with different concentration were all significantly different ( all P 〈 0, 01). While under the same concentration, the cell survival rate was also significantly different with different incubating time except that the concentration was 0 μg/ml and 0. 1 μg/ml ( all P 〈 0.05 ). Conclusion Under the special light source,the phortofrin incubating concentration and incubating time significantly influence photodynamic therapy effect in human esophageal cancer cell line in vitro.
