摘要
目的探讨mtr系统若干基因变异与淋球菌对阿奇霉素耐药性的相关性。方法对临床分离的淋球菌阿奇霉素耐药性代表株mtrR基因序列及mtrR启动子区域回文结构序列的突变进行分析,使用荧光定量PCR技术对结构基因mtrC的表达水平进行测定。结果所有中等以上耐药菌株均发生mtrR基因H105Y的变异,伴随mtrR启动子区域回文结构的缺失突变;敏感株有一株出现G45D的突变,mtrR启动子区域回文结构未检测出突变;中等以上耐药菌株与敏感菌株相比mtrC基因表达有显著差异(P<0.05)。结论mtrR基因的H105Y变异和回文序列碱基缺失与mtrC表达有负向相关关系,与淋球菌阿奇霉素耐药性显著相关。
Objective Exploring some mutations within mtr Systen with the Azithromycin-resistance of Neisseria gonorrhoeae. Methods MtrR gene was amplified by PCR reactions, then sequenced. Real-time PCR was used to measure the expression level of mtrC gene. Results M1 of Azithromycin-intermediate and resistant strains except from those sensitive trains, had been detected an H105Y mutation within mtrR gene, and a base "A" deletion within its promoter region ( P 〈 0.05 ). The expression levef of mtrC was different among three groups ( sensitive, intermediate and reisistant, P 〈 0.01 ). Condusion H105Y mutation and the base-A deletion in mtrR negatively correlated with the expression of mtrC geness. Those mutations correlated with the resistance to Azithromycin.
