摘要
目的在基因及蛋白水平分析αvβ6整合素在子宫内膜异位症(endometriosis,EM)的在位内膜、异位内膜及对照组正常在位子宫内膜的表达状况,探讨其表达的生物学意义。方法用半定量逆转录-聚合酶链反应(RT-PCR)、免疫组化法对34例EM患者在位和异位内膜,28例非EM患者的在位子宫内膜αvβ6整合素的表达进行半定量、定位分析,并与EM临床分期、类型及部位相联系。结果αvβ6整合素基因和蛋白在EM患者异位内膜、在位内膜及对照组正常在位内膜的腺上皮中均有不同程度的表达。异位内膜αvβ6整合素mRNA表达显著高于EM在位内膜(P<0.01);EM在位内膜αvβ6整合素mRNA表达显著高于对照组正常在位内膜(P<0.01)。EM异位内膜αvβ6蛋白质阳性表达率高于在位内膜(P<0.05),EM在位内膜阳性表达率高于对照组正常在位内膜(P<0.05)。卵巢子宫异位囊肿壁αvβ6整合素mRNA值与腹膜红色样病变相比差异无统计学意义(P>0.05),2组与宫骶韧带结节αvβ6整合素mRNA值相比差异有统计学意义(P<0.01)。增殖期与分泌期在位内膜αvβ6整合素基因及蛋白的表达比较差异无统计学意义(P>0.05)。不同期别αvβ6基因及蛋白表达比较差异无统计学意义(P>0.05)。结论αvβ6整合素参与了EM的发病过程,不同部位异位内膜的αvβ6整合素表达量有显著不同。
Objective To explore the biological significance of αvβ6 integrin expression in ectopic and eutopic endornetrium in patients with endornetriosis（EM）. Methods Reverse transcriptase polymerase chain reation （RT-PCR） and immunohistochenfistry were used to examine the expression of αvβ6 integrin in 34 patients with endornetriosis and 28 cases of eutopic endornetrium of the control group. Results All glandular epithelial cells of eutopic and ectopic endornetriurn in endornetriosis and eutopic endornetriurn in the normal control expressed αvβ6, in different degree. The gene expression of αvβ6 in eutopic endornetrium of EM group was significantly higher than that of eutopic endornetrium of the control group （ P 〈 0.01 ）. The gene expression of αvβ6 in ectopic endornetrium was significantly higher than that of eutopic endornetrium of EM（ P 〈 0.01 ）. The positive percentage of αvβ6, integrin in eutopic endornetrium of EM group was significantly higher than that in endornetriurn of the control group （ P 〈 0.05）. The positive percentage of αvβ6 integrin in eetopie endornetriurn was significantly higher than that in eutopic endornetrium of EM, （ P 〈 0.05）. There was no significant difference between the gene expression of αvβ6 integrin of ovarian endornetrioma and red peritoneal lesion, and both of them were significantly higher than uterosacral ligament nodule（ P 〈 0.01 ）. The expression of αvβ6 integrin gene and protein have no significant difference （ secretory versus proliferative） （ P 〉 0.05 ）. No significant difference was observed between early stage and advanced stage of EM in both levels of protein and gene （ P 〉 0.05）, Conclusion The up-regulated expression of αvβ6 integrin in eutopic and eetopie endornetium in EM suggests that αvβ6 integrin may play a role in the development of EM.
