摘要
目的探讨补气活血中药治疗溃疡病的分子生物学机制。方法将126只Wistar大鼠随机分为正常组、模型组、当归组、丹参组、黄芪组、党参组、洛赛克组。除正常组外,其余各组采用Okabe乙酸涂抹法制作大鼠实验性胃溃疡模型,在造模结束后24 h,予相应药物干预。在第7、14、21天分批处死大鼠,剖腹取胃;观测溃疡愈合指数,光学显微镜下观察溃疡愈合质量,免疫组化方法检测表皮生长因子受体(EGFR)和碱性成纤维细胞生长因子受体(bFGFR)蛋白的表达情况。结果当归、丹参、党参、黄芪均可以促进溃疡愈合,其愈合质量优于洛赛克组,其中活血药当归、丹参组溃疡底部瘢痕组织较成熟,补气药党参、黄芪组新生黏膜上皮接近正常。溃疡形成后造模各组EGFR、bFGFR表达增加,并且随着溃疡愈合其表达逐渐减弱,在溃疡愈合的各个阶段,当归、丹参组中bFGFR表达水平均优于其他组别(P<0.05),而党参、黄芪组中EGFR的表达水平高于其他组别(P<0.05),洛赛克组则与模型组差异不显著。结论当归、丹参、党参、黄芪均能够促进溃疡愈合;补气药对EGFR的作用显著,活血药对bFGFR的作用明显,其差异性可能与其在EGFR和bFGFR表达上的作用不同有关。
Objective This study aimed to investigate the molecular biological mechanism of Chinese herbs. Methods 108 rats were randomly divided into six groups, including normal, model, Losec, Danggui, Dansheng, Dangshen and Huangqi groups. Gastric ulcer was produced in rats by serosal application of acetic acid. Rats were gavaged once daily with 1ml/100g body weight of Dangshen, Huangqi, Danggui and Danshen 5g/kg, Losec 20mg/kg, or normal saline starting 24h after ulcer induction. On days 7, 14, 21 after treatment, rats were anesthetized with nembutal and stomachs were removed for different detection. We determined the gastric ulcer index(GUI) and histological assessment of quality of healing ulcer, and examined the gastric mucosal expressions of EGFR and bFGFR. Results Each of the herb Dangshen, Huangqi, Danggui and Danshen could improve the healing of gastric ulcer; expressions of EGFR and bFGFR were enhanced after the ulceration, while expressions of bFGFR in Danggui and Danshen groups at different phases were the highest (P〈0.05 versus each group). On the other hand, expressions of EGFR in Dangshen and Huangqi groups were the highest (P〈0.05 versus each group). There were no significant differences compared model group with losec group at different phases. Conclusion Dangshen, Huangqi, Danggui and Dansben can significantly reduce the area of experimental gastric ulcer;, Dangsben and Huangqi can significantly improve the expression of EGFR; Danshen and Danggui improve the expression of bFGFR significantly; the differences between qi-enriching and blood-activating herbs on these factors depend upon their effects on the expressions of EGFR and bFGFR.
