三氧化二砷对类风湿关节炎滑膜细胞增殖及分泌作用的影响 被引量:2

摘要 目的观察三氧化二砷(As2O3)对体外培养的RA滑膜成纤维细胞的增殖及分泌功能的影响。方法体外培养RA滑膜成纤维细胞,用含As2O3浓度分别为0、0.5、2、5μmol/L的培养液分别作用于培养细胞,24 h后检测上清液TNF-α和IL-1的水平;采用TUNEL法和流式细胞仪测定细胞增殖情况。结果As2O3作用组TNF-α及IL-1水平均较对照组降低(P<0.05),随浓度梯度的增加作用增强。TUNEL法检测细胞增殖,As2O3作用第3天后各实验组均明显低于对照组(P<0.05),且其作用呈剂量依赖性。滑膜细胞细胞周期在As2O3作用组均与对照组有明显差异,其变化亦呈剂量依赖性。结论As2O3可促进RA滑膜B型细胞增殖,抑制TNF-α及IL-1的表达。 Objective To observe the effect of arsenic trioxide ( As2O3) on synovlocytes' proliferation and cytokines excretion in rheumatoid arthritis (RA). Methods Synoviocytes of RA were cultured in vitro. Different concentrations(0, 0, 5, 2 and 5 umol/L) of As2O3 were applied in the cultured concentrations. The concentrations of TNF-a and IL-1 were observed 24 hours later. TUNEL and flow cytometry were used to detect the proliferation of synoviocyte. Results Compared with the control group, the concentrations of TNF-a and IL-1 decreased significantly in As2O3 group ( P 〈 0.05 ). The effects were more apparent when the concentrations of As2O3 were increased. Cell proliferation were significantly decreased in As2O3 group compared with that in control group after three days ( P 〈 0.05 ), and the effect was concentration-depend- ent. The cell cycles of synoviocyte were significantly different between As2O3 group and the control group ( P 〈 0.05 ), and the effects showed concentration-dependent. Conclusions As2O3 enhances the proliferation of synoviocyte but inhibits the expressions of secretion of TNF-a and IL-1.
