摘要 目的:探讨γ干扰素(IFN-γ)对低密度脂蛋白免疫复合物(LDL-IC)诱导U937细胞基质金属蛋白酶-1(MMP-1)表达的影响。方法:采用不同浓度的LDL-IC诱导不同浓度的IFN-γ预处理的U937细胞,通过RT-PCR分析MMP-1 mRNA表达水平的变化。结果:正常U937细胞MMP-1表达量低。LDL-IC可增强U937细胞MMP-1mRNA的表达,而单独IFN-γ处理对MMP-1表达无影响;LDL-IC对经IFN-γ预处理的U937细胞作用效果较未经处理的更强(P<0.01),并具剂量效应。结论:IFN-γ可增加LDL-IC诱导的U937细胞MMP-1表达,加速动脉粥样硬化的发生、发展。 Objective : To investigate the effect of pretreatment of U937 cells with interferon-γ (IFN-γ) on the matrix metalloproteinase-1 (MMP-1) expression induced by low- density lipoprotein (LDL) immune complexes (IC). Methods: U937 cells pretreated with IFN-γ were incubated with LDL-IC, and the mRNA expression of MMP-1 was measured by reverse transcriptase polymerase chain reaction (PT- PCR). Results: MMP-1 was lowly expressed in the normal U937 cells. LDL-IC significantly increased the MMP-1 mRNA expression, while simple IFN-γ did not directly stimulate the MMP-1 expression in the U937 cells. LDL-IC induced a higher MMP-1 expression in the U937 cells pretreated with IFN-γ than in the untreated ones (P 〈 0. 01 ). Conclusion: IFN-γ pretreatment enhances the LDL-IC induced MMP-1 expression in U937 cells.
