摘要
目的研究中华补血草根提取物(the extract of Limonium sinense Ktze root,LSE)对小鼠急性肝损伤的防护作用及药理机制。方法建立小鼠D-氨基半乳糖/脂多糖(D-GalN/LPS)肝损伤模型,检测LSE对血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)活性的影响;观察肝脏组织超微结构的变化;测定肝线粒体膜电位以及线粒体对高钙诱发肿胀的敏感性的变化。结果一定剂量(100~400mg·kg^-1)LSE能有效抑制D-GalN/LPS引起的小鼠血清AST及ALT活性的显著上升;明显减轻肝损伤引起的小鼠肝脏组织超微结构的破坏;抑制线粒体膜电位的降低以及线粒体对外加高钙诱发肿胀敏感性的下降。结论LSE对D-GalN/LPS诱导的急性肝损伤具显著的防护作用,其机理可能与保护肝脏线粒体、抑制肝细胞凋亡有关。
Objective To study the hepatoprotective effect of the extract of Limonium sinense (Girard) Ktze root (LSE) on acute liver injury induced by D-galactosamine and lipopolysaccharide (D-GalN/LPS) and its mechanism. Methods D-GalN/LPS -induced liver injury model was used to evaluate the effect of LSE on the activities of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in mice. Uhrastructure of liver was observed and liver mitochondrial membrane potential and mitochondrial swelling were measured following D-GalN + LPS injection without or with LSE. Results 100 mg·kg^-1 , 200 mg·kg^-1 or 400 mg·kg^-1 LSE could significantly inhibit the increase of serum AST and ALT activities induced by D-GalN/LPS improve morphological changes in liver and inhibit the decrease of mitochondrial membrane potential and sensitivity of mitochondrial swelling to the exotic Ca^2+ stimulation. Conclusion LSE has hepatoprotective activity and the mechanisms may be related to its protection on liver mitochondria.
