摘要
以工程菌PP-MS-NPm-4-16为发酵菌株,探讨其在5L发酵罐上发酵产植酸酶的条件,从而为植酸酶规模化生产提供理论依据。对发酵过程中的补料方式、甲醇流速、甘油流速及诱导时间进行了分析。通过正交试验筛选出工程菌产植酸酶的最佳发酵条件。在温度28℃、搅拌速度750r/min条件下,接种量10%,恒溶氧补料,甲醇流速8mL/(L·h),诱导阶段甘油流速1mL/(L·h),诱导时间84h,酶活可以达到472.933kU/mL发酵液。
In order to provide theoretical basis for the industrial production of phytase, the phytase fermentation conditions of the genetic-engineered Pichia pastoris strain PP-MS-NP^m-4-16 in the 5 L fermentation cylinder were studied. The fed-batch way, methanol flow rate, glycerol flow rate and induce time were analyzed. The optimum condition of phytase fermentation was determined by using orthogonal design. The result indicated that the temperature 28 ℃, stirring speed 750 r/min, inoculum size 10%, methanol flow rate 8 mL/(L·h), glycerol flow rate during inducing phase 1 mL/ (L·h),induce time 84h, and the phytase activity was reached to 472.933 kU/mL.
