摘要
目的:研究环氧化酶-2(COX-2)在早期糖尿病大鼠视网膜中的表达,以探讨其在糖尿病视网膜病变发生和发展中的作用。方法:将140只SD大鼠随机分为两组,对照组(N组)与糖尿病组(DM组),DM组采用链脲佐菌素(STZ)一次性尾静脉注射建立糖尿病模型。应用PV免疫组织化学染色分别于1、2、4、8、12周检测视网膜中COX-2蛋白的表达。结果:COX-2在正常对照组大鼠视网膜基本不表达,糖尿病模型诱导成功后1周,COX-2于内核层有弱表达(P<0.05),随时间的延长表达量逐渐增高,至糖尿病12周(DM12)COX-2的表达量约为糖尿病1周(DM1)的5倍(P<0.05)。结论:COX-2在早期糖尿病视网膜细胞中呈高表达,可能在糖尿病视网膜病变发生和发展中起到重要的作用。
Objective: To detect the expression of cyclooxygenase-2 in the retina of diabetic rats ,and investigate the role of it in the pathogenesis of diabetic retinopathy (DR).Methods:140 SD rats were randomly divided into two groups- control group (N)and diabetes mellitus group (DM). Diabetes mellitus was induced by streptozotocin (STZ) vena caudalis injection in DM group. PV Immunohis-tochemical staining was taken to detect the expression of COX-2 in retina respectively at 1,2,4,8, 12 weeks after STZ injection. Results: The result of immunohistochemistry showed that the expression of COX-2 protein in retina was not found in control group basically, but was found in the diabetic retina as early as 1 week after induction of diabetes and it mainly localized in the inner nuclear (P〈0.05). The longer duration of the disease was, the more expression of COX-2 was. The expression in DM12 was 5 fold compared with DM1 (P〈0.05). Conclusion: Abnormal expression of COX-2 in the retina of early stage diabetic retinopathy plays an important role in the pathogenesis of DR.
