摘要
分析了不同DNA浓度、Mg2+、dNTP、引物用量对朱砂根ISSR-PCR扩增结果的影响,确立适合朱砂根的ISSR反应体系为:在20μl反应体系中,含1×buffer、30ng模板DNA、随机引物量0.4μmol/L、dNTP0.15 mmol/L、Mg2+1.75 mmol/L、1UTaq DNA聚合酶。
To explore the optimum ISSR reaction condition in Ardisia crenata,the effect of different concentrations of template DNA,Mg^2+ and dNTP etc.on amplification results were analyzed in this study.Results showed that the optimum reaction condition in a total volume of 20μl included the following components: 1×buffer,template DNA of 30 ng,primer of 0.4μmol/L,dNTP of 0.15mmol/L,Mg^2+ of 1.75mmol/L and Taq polymerase of 1U.
