摘要
目的观察趋化肽fMLP与抗肿瘤抗生素博安霉素(BAM)序贯给药对肿瘤生长的影响。方法采用小鼠肝癌H22移植瘤模型观察二者序贯给药的作用。结果fMLP 1 mg/kg瘤周给药×3,对肿瘤的生长没有影响。BAM 20 mg/kg腹腔给药×1,设计了以下两个实验:(1)先给BAM后给fMLP,合用组肿瘤生长速度与单用BAM组的相似,BAM组在14 d的抑瘤率58.9%,合用组为51.1%,CDI>1;(2)先给fMLP后给BAM,合用组肿瘤生长速度比单用BAM组明显减慢,BAM组在14 d的抑瘤率28.4%,合用组为51.3%,P<0.05,CDI=0.73。结论先给趋化肽fMLP后给BAM对小鼠肝癌H22移植瘤的生长抑制有协同作用。
Objective This study is set to investigate the antitumor activity of fMLP in sequential combination with (BAM). Methods The model of subcutaneously transplanted heptoma 22 in mice was used to evaluate the therapeutic effect. Results In all in vivo experiments, fMLP was administered peritumorally at the dose of 1 mg/mouse; no significant inhibition by fMLP alone on the growth of hepatoma 22 was found. Different settings of BAM and fMLP combination included: 1) BAM was once administered ip on day 4 after tumor inoculation, fMLP was administered peritumorally on day 7, day 10 and day 14 after tumor inoculation. BAM (20 mg/kg) alone and BAM-fMLP sequential combination inhibited the growth of hepatoma 22 by 58.9% and 51.1%, respectively (P〉0.05, CDI〉I) on day 14. fMLP was administered peritumorally on day 1, day 3 and day 5 after tumor inoculation. BAM (20 mg/kg) was once administered ip on day 7 after tumor inoculation. The growth of tumor in BAM (20 mg/kg) group was faster than that in fMLP-BAM sequential combination group. On day 14, BAM (20 mg/kg) alone and fMLP-BAM sequential combination suppressed the growth of tumor by 28.4% and 51. 3%, respectively (P〈0.05, CDI=0. 73). Conclusion The antitumor effect of fMLP-BAM sequential combination shows synergism when fMLP prior to BAM is administered.
