摘要
目的:通过观察电针对脑缺氧缺血新生大鼠海马结构中神经上皮干细胞蛋白(Nestin)与5-溴-2-脱氧脲嘧啶(Brdu)表达的影响,了解电针对脑缺氧缺血后神经发生水平的干预,为电针穴位疗法的临床应用提供实验依据。方法:100只7日龄健康Wistar大鼠,体重12~15g,随机分为4组:空白对照组、模型判定组、缺氧缺血组与缺氧缺血后电针治疗组,各组动物(除空白对照组外)分别结扎左侧颈总动脉,然后将动物置于8%O2-92%N2混合气中2h后取出。模型判定组动物在缺氧缺血48h后处死,制备脑组织切片。脑缺氧缺血后电针组在缺氧缺血后恢复2d,开始刺激"百会"和"大椎"两穴。24d后除模型判定组外,各组动物分别取左侧脑组织制作标本,行Nestin与Brdu免疫组化染色。结果:脑缺氧缺血后电针组海马CA1区Nestin免疫阳性细胞数高于正常对照组及脑缺氧缺血组(P<0.05)。脑缺氧缺血后电针组齿状回Brdu免疫阳性细胞数高于正常对照组及高于脑缺氧缺血组(P<0.05)。结论:电针穴位治疗可明显提高(加强)脑缺氧缺血后海马结构神经发生的水平(能力)。
Objective: To observe the influence of electroacupuncturing on the Nestin and Brdu expressions in the hippoeampal formation of neonatal rats with hypoxia-isehemia, and to confirm the effect of eleetroaeupueture(EA) on HIBD (Hypoxie-isehemie brain damage) in neonatal rats, and then provide the theoretical basis for clinical application. Methods:7 days old Wistar rats (n=100), weighing 12-15 g, were randomly divided into four groups: the control group, the model group, HIBD group and electroacupucture(EA) group. Except the control group, the other three groups were prepared by left common carotied artery ligation and systemic hypoxia (8% oxygen in nitrogen) for 2 h. Then the rats in model group were sacrificed. After 2 d of recovery, the rats in electroacupucture group were treated with electroacupucture ("baihui" and "dazhui"points) every day till the rats were sacrificed 22 d after HIBD. The rest rats were subjected to perfusion-fixation 22 d later, the left brains were regularly paraffin wax embedded, sectioned and stained by Nestin and Brdu immuno- histochemical staining. Results: Nestin-positive cells distributed widely in the hippocampal CA1 ,CA2,CA3 and dentate gyrus of hippocampus formation in each neonatal rat brain. The number of Nestin-positive ceils in the electroacupucture group increased markedly, as compared with normal control group and HIBD group. The Brdu-positive ceils were mainly located in subventricular zone of lateral wall, subgranular zone of dentate gyrus and corpus callosum in each neonatal rat brain. In the rats of EA group, the Brdu-positive cells were conspicuously higher than those in the normal control group. Conclusion: Eectroacupucture compared with hypoxia-ischemia gives conspicuously rise to enhance neurogenesis in the hippocampal formation of neonatal rats after hypoxia-ischemia.
