摘要
目的:分析卵巢癌组织中白细胞介素18(IL-18)及白细胞介素1β转化酶(ICE)的基因及蛋白表达水平,探讨其与卵巢癌临床特性之间的关系。方法:分别采用RT-PCR及免疫组化染色法检测正常卵巢(n=10)、卵巢良性肿瘤(n=6)、卵巢癌(n=32)组织中IL-18、ICE的mRNA及蛋白表达;分析卵巢癌不同病理类型、临床分期及发病年龄患者之间表达是否具有差异。结果:①正常、良性肿瘤及卵巢癌组织中均有IL-18mRNA表达(分别为1.30±0.48、1.02±0.17、0.77±0.35);ICEmRNA在正常及良性肿瘤组织中均有表达,卵巢癌组织中有29例(90.63%)表达(分别为0.96±0.51、0.73±0.34、0.63±0.34)。②IL-18及ICE蛋白均表达于卵巢上皮细胞浆内。正常、良性肿瘤及卵巢癌组织的IL-18阳性表达率分别为83.3%、80.0%和31.3%;ICE阳性表达率分别为80.0%、50.0%和37.5%。③卵巢癌组织中IL-18、ICEmRNA及蛋白表达均较正常及良性肿瘤组织明显减少(均P<0.05),良性与正常组织之间相比差异均无显著性(均P>0.05)。卵巢癌组织中IL-18与ICE的mRNA、蛋白表达之间呈显著正相关(分别为r=0.37,P=0.036;r=0.45,P=0.009)。④IL-18、ICEmRNA及蛋白表达在不同病理类型、临床分期及发病年龄患者之间均未见显著性差异(均P>0.05)。结论:卵巢癌患者局部癌组织IL-18、ICE表达减弱,可能与卵巢癌的发生发展有着一定的关系。
Objective: To study the gene and protein expressions of interleukin 18 (IL-18) and interleukin 1-β converting enzyme (ICE) in human ovarian cancer tissues, and to analyze the relationship between the expressions and clinical characteristics. Methods: The mRNA and protein levels of IL- 18 and ICE in local ovarian tissues from normal ( n = 10) ,benign tumor ( n = 6) and epithelial cancer group ( n = 32) were determined by RT-PCR and immunohistochemistry staining, respectively. The expression diversity among different pathological types, clinical stages and ages was analyzed.Results:①The IL-18 mRNA was expressed in all the normal,benign tumor and ovarian cancer tissues ( 1.30 ± 0.48,1.02 ± 0.17,0.77 ± 0.35, respectively) ; ICE mRNA was expressed in all the normal and benign tumor tissues, but expressed in 29 of 32 (90.63 % ) ovarian cancer tissues (0.96-± 0.51,0.73 ± 0.34,0.63 ± 0.34, respectively). ②The protein staining of IL-18 and ICE was in cytoplasm of ovarian epithelial cells. The percentages of IL-18 positive expression in normal, benign tumor and cancer tissues were 83.3 %, 80.0%, 31.3 %, respectively; And those of ICE were 80.0 %, 50.0 %, 37.5 %, respectively. ③The gene and protein levels of IL-18 and ICE in ovarian cancer tissue were lower than those of normal and benign tumor tissues (all P 〈 0.05) with no significant difference between normal and tumor tissues ( P 〉 0: 05) with each other (all P 〉 0.05). There were positive correlations in both gene and protein level between IL- 18 and ICE ( r = 0.37, P = 0. 036; r = 0.45, P = 0.009, respectively).④ Significant changes of IL- 18 and ICE expressions were not detected among different pathologic types, clinical stages and ages of patients with ovarian cancer ( all P 〉 0.05). Conclusion: The decreased expressions of IL-18 and ICE in local ovarian cancer Tissues should have certain relation to the occurrence and development of ovarian cancer.
