摘要
[目的]研究谷氨酰胺、表皮生长因子对体外受精胚8细胞以后发育的影响。[方法]通过对牛进行体外受精和受精卵体外培养,研究谷氨酰胺、EGF对牛胚胎体外发育的影响。[结果]添加谷氨酰胺的4-8细胞发育率和桑囊率(52.52%、30.22%),都比对照组高,但差异不显著(P〉0.05)。联合添加谷氨酰胺和表皮生长因子的4-8细胞发育率(55.03%)比对照组高,但差异不显著(P〉0.05);桑囊率(32.39%)比对照组(27.46%)高4.93百分点,且二者之间差异显著(P〈0.05)。联合添加谷氨酰胺和表皮生长因子组比单独添加谷氨酰胺组的4-8细胞发育率和桑囊率都高,但差异不显著(P〉0.05)。[结论]单独添加谷氨酰胺对早期胚胎的体外发育有一定的促进作用,联合添加谷氨酰胺和表皮生长因子对帮助牛胚胎通过体外发育阻滞期有良好的促进作用。
[ Objective ] The research aimed to study the effects of glutamine and epidermal growth factor (EGF) on the growth of in vitro fertilization embryos (after 8- cell). [Method] Though in vitro fertilization and zygote culture in vitro, the effects of glutamine and EGF on the growth of bovine embryos in vitro were studied. [Result] The 4 - 8 cell growth rate mad mulberry vesicle rate with adding glutamine were 52.52% and 30.22% respectively, being higher than that in control group and having no significant difference ( P 〉 0.05). The 4 - 8 cell growth rate with adding glutamine and EGF was 55.03%, being higher than that in control group and having no significant difference (P 〉 0.05). And the mnlberry vesicle rate was 32.39%, being higher 4.93 percentage points than that in control group and having significant difference ( P 〈 0.05). The 4 - 8 cell growth rate and mulberry vesicle rate in the group of adding glutamine and EGF were higher than that in the group of only adding glutamine, but the difference was not significant ( P 〉0.05). [Conclusion] Adding glutamine singly had certain promotion on the in vitro growth of early embryos. And adding glutamine and EGF had good promotion to help bovine embryo to pass the development block period in vitro.
