摘要
研究了用高效液相色谱法测定茶叶样品中的6种茶多酚。6种茶多酚用70%甲醇-水溶液提取,以Agilent ZORBAX SB-C18液相色谱柱(4.6 mm×250 mm,5μm)色谱柱为固定相,0.5%醋酸水溶液和甲醇梯度洗脱为流动相,茶叶中主要的茶多酚均达到基线分离;用紫外二极管矩阵检测器检测,得出各组分在其最大吸收波长下的色谱图,根据该色谱图的峰面积定量,并用紫外光谱图对茶叶中主要多酚进行辅助定性。标准回收率为92.5%-102%;RSD为1.2%-1.5%。
Determination of 6 polyphenols from tea by high performance liquid chromatography was studied. The poly- phenols was extracted from tea sample by refluxing with 70% methanol, and separated on a Zorbax SB - C18 chromatographic column (4.6 mm× 250 mm, 5μm) eluting with gradient of methanol and 0.5 % acetic acid solution at a flow rate of 1 mL/ min, and monitored by photediode array detector at maximum absorption wavelength. Six polyphcnols from tea sample could be simultaneously determined with recovery of 92.5% - 102% and relative standard deviation of 1.2% - 1.5%.
