摘要
目的运用高胰岛素-正常血糖钳夹实验来评价桑叶黄酮提取物对高脂喂养的2型糖尿病大鼠胰岛素抵抗的影响。方法高脂(脂肪占热量56.89%)喂养成年雄性SD大鼠8周诱导大鼠胰岛素抵抗,一次性腹腔注射链脲菌素(STZ,25mg/kg),1周后测空腹血糖≥200mg/dl纳入实验,大鼠随机分为4组:糖尿病模型组、罗格列酮治疗组、桑叶黄酮治疗组和对照组。8周后运用高胰岛素-正常血糖钳夹实验测GIR,并取血测血糖、血脂、PPARγ以及胰岛素。结果桑叶黄酮治疗组GIRI、SI和PPARγ明显高于糖尿病组(P<0.05),而血脂明显降低(P<0.05)。结论桑叶黄酮能增加高脂喂养2型糖尿病大鼠胰岛素敏感性,同时具有降低血脂的作用。
Objective To establish an animal model and evaluate mulberry leaves flavonoids( MTF ) on improve insulin resistance in rat of type 2 diabetes with hyperinsuliuemic-euglycemic clamp. Methods Adult male SD rats were red with high-fatty diet for 8 weeks to induce insulin resistance, then streptozotion (STZ, 25mg/kg) was injected intraperitoneaUy to result in insulin secretion defect and hyperglycemia. The rats whose fasting blood glucose levels ≥ 200mg/all were entered into the study after injected STZ 1 wk. the blank control group given ordinary diet and the positive control group given high-fat diet were injected 5ml/kg of normal sodium intragastrically. rosiglilazone group was given rosiglitazone while MTF group was given mulberry leaves flavonoids solution was injected iutragastrically. They all were given administration once a day for eight weeks. Insulin resistance indexes were measured using the hyperinsulincmic euglycemic glucose clamp technique before sacrifice. Results GIR, PPARγ and ISI of MTF group are significantly higher than those of B group( P 〈 0.05 ). ConcIusion MTF could have effect in increasing insulin sensitivity and reducing serum lipids.
