厄贝沙坦对实验性糖尿病大鼠肾脏的保护作用 被引量:1

摘要 目的探讨厄贝沙坦对实验性糖尿病大鼠肾脏的保护作用及其可能机制。方法Wistar健康大鼠24只,随机分为对照组、模型组、药物组,每组8只。链脲佐菌素诱导糖尿病大鼠模型,8周后抽血测血糖(BG)、糖化血红蛋白(HbA1c)、尿素氮(BUN)和肌酐(SCr),计算尿白蛋白分泌率(UAER)。用免疫组化检测大鼠肾脏细胞间黏附分子1(ICAM-1),Western blot检测血管细胞黏附分子1(VCAM-1)蛋白表达水平。结果模型组与药物组的BG、HbA1c、BUN、SCr都高于对照组(P<0.05)。模型组UAER高于对照组,而药物组则低于模型组(P<0.05)。药物组ICAM-1与VCAM-1蛋白表达量低于模型组(P<0.05)。结论厄贝沙坦对实验性糖尿病大鼠的肾脏有保护作用,可能与抑制ICAM-1与VCAM-1在肾脏的表达有关。 Objective To investigate the protective effects of irbesartan on the kidneys of diabetic rats, and the possible mechanisms. Methods Twenty-four male wistar rats were randomly and equally divided into control group, model group and treatment group. The rat model of DM was induced by injection of streptozocin (STZ). Eight weeks later, blood glucose(BG), glycosylated hemoglobin lc (HbAlc), blood urea nitrogen(BUN), serum creatinine (SCr) and urinc albumin excretion rate (UAER) were measured. The expression of intercelluar adhesion molecule-1 (ICAM-1) was observed by immunohistochemistry. The expression of vascular cell adhesion molecule-1 (VCAM-1) was observed by Western blot. Results The levels of BG, HbAlc, BUN and SCr in model group were higher than those in control group (P〈0.05). UAER in model group was higher than that in control group and treatment group. The expressions of ICAM-1 and VCAM-1 in treatment group were decreased significantly compared with those in model group. Conclusion Irbesartan seems to have protective effects on the kidney of diabetic rats. The effects may be achieved by decreasing the expressions of ICAM-1 and VCAM-1.
