摘要
目的建立检测人血浆盐酸伪麻黄碱的HPLC法。方法以ZORBAX SB-C 18为色谱柱;乙腈-0.15 mol·L^(-1)磷酸二氢钠(pH=3.0)-水-0.5%十二烷基磺酸钠(SDS)(33:15:42:10)作流动相,流速1.0mL·min^(-1);血浆样品经乙酸乙酯和正己烷提取后,有机相用盐酸酸化,下层水相在206 nm波长下进行检测。结果盐酸伪麻黄碱血浆浓度在5.0~400μg·L^(-1)时线性关系良好(r=0.999 9),最低检测浓度为5.0μg·L^(-1);低、中、高浓度的日间、日内RSD(n=5)均<10%;相对回收率均在85%~115%内,绝对回收率>75%。结论本方法操作简便,结果准确,适用于血浆中盐酸伪麻黄碱浓度的检测。
AIM To establish a HPLC method for the determination of pseudoephedrine hydrochloride in human plasma. METHODS The analytical column was packed with ZORBAX SB - C18. The mobile phase was aeetonitrile- 0.15 mol·L^-1 NaH2PO4(pH = 3.0)- water-0.5%SDS (33: 15:42: 10) .The flow rate was 1.0 mL·min^-1. The detection wavelength was 206 nm. Plasma samples were extracted with ethyl acetate and N-hexane. The organic phase was separated and acidified with 0.1 mL 1.5 % HCl, and detected at 206 nm. RESULTS The liner range of pseudoephedrine hydrochloride in human plasma was 5.0- 400μg·L^-1( r = 0. 999 9). The limit of detection was 5.0μg·L^-1. Intra-day and inter-day RSD for assaying the plasma sample containing three concentration of pseudoephedrine hydroehloride were both lower than 10%. Relative recovery rate was at 85% - 115% and absolute recovery rate was more than 75%. CONCLUSION The HPLC method to determine pseudoephedrine hydroehloride in plasma is simple, high selectivity and sensitivity.
