摘要
目的检测缺氧缺血性脑损伤(hypoxic-ischemic brain damage,HIBD)后Foxg1基因及脑皮质特定基因(Foxp2,ER81,Otx1,Foxp1,RORβ,Reelin)的动态变化。方法采用Rice法制作新生鼠HIBD动物模型,假手术组作为对照组,分别在术后第3、7、14、28、56d取右侧脑半球。半定量RT-PCR检测各基因表达水平。结果HIBD后Foxg1基因表达水平模型组3d较对照组低(P<0.05),14、56d表达水平均较对照组高(P<0.05);Reelin基因表达水平在术后7d及28d均较对照组高(P<0.05);Foxp2基因在7、14、56d表达水平均较对照组高(P均<0.05)。Otx-1基因在术后7、14、28、56d表达水平均较对照组高(P均<0.05),模型组与对照组Otx-1的表达均呈下降趋势;RORβ、Foxp1、ER81表达趋势尚不能肯定。结论HIBD后Foxg1、Reelin及Foxp2表达水平呈上升趋势,Otx1呈下调趋势,RORβ、Foxp1、ER81表达趋势尚不能肯定,Foxg1基因表达变化与脑损伤后大脑结构自身修复的具体机制有待进一步研究。
Objective To observe the alteration of Foxg1, Foxp2, ER81, Otx1, Foxp1, RORβ, Reelin genes expressing in cerebral cortex in neonatal rat during hypoxic ischemic brain injury (HIBD). Methods The HIBD model was established according to Rice method. The expression of each gene in the right cerebral hemisphere was examined by semi-quantitative RT-PCR on 3^rd, 7^th, 14^th, 28^th and 56^th day respectively. Results Compared with the control group, the expression level of Foxg1 in HIBD group was lower on 3^rd day, higher on 7^th and 28^th day ( P 〈 0. 05 ), that of Reelin was higher on 3^rd, 14^th and 56^th day ( P 〈 0.05 ), that of Foxp2 was higher on 7^th, 14^th and 56^th day (P 〈0.05) , and that of Otx-1 was higher on 7^th, 14^th, 28^th and 56^th day (P 〈0.05). The expression level of Otx-1 was down-regulated in both HIBD and control groups, whereas the expressions of RORβ, Foxp1 and ER81 in this experiment were difficult to determine. Conclusion The expression levels of Foxg1, Reelin and Foxp2 were up-regulated, but the expression level of Otx1 was down-reg- ulated, while the alteration tendency of RORβ, Foxp1 and ER81 were unclear in this experiment. The relationship between Foxg1 gene and repair of brain structure will be explored in future.
