摘要 将培养到根长至1 cm左右的大豆根浸入到浓度分别都为50 mmol/L的、以NO3-为背景的Ca2+、Mg2+混合溶液中,在处理到48 h和60 h时进行细胞形态学观察、TUNEL检测、单细胞凝胶电泳以及丙二醛含量和细胞膜透性的测定,将浸入去离子水中培养的作为对照,再将浸入浓度都是100 mmol/L、以NO3-为背景的Ca2+溶液中和Mg2+溶液中,分别处理48 h进行TUNEL检测。试验结果表明经Ca2+、Mg2+单独处理的大豆根没有发生细胞程序性死亡(PCD),而Ca2+、Mg2+混合处理的大豆根发生了PCD。所以大豆根产生PCD是Ca2+、Mg2+共同诱导的结果,并推测了其发生PCD的机制。 Roots of the soybeans,that grew about 1cm,were immerged into the solution whose concentration of Ca2+ and the Mg2+ were 50mM taking NO3-as background for 48h and 60h for observing the morphostructure of the soybean root cells,TUNEL assay,single cell gel electrophoresis and mensuration of the malondiadehyde content and cell membrane permeability of the soybean root cells.Roots of the soybeans were immerged into the water without ion for comparison,and were separately immerged into the contents whose concentration of Ca2+ was 100mM taking NO3-as background and the same contents of Mg2+ for 48h for TUNEL assay.The results of experiments indicated that Roots of the soybeans treated by the solution of Ca2+ and Mg2+ together produced programmed cell death(PCD),while Roots treated by the solution of Ca2+ and the contents of Mg2+ individually did not produce PCD.According to this result,we believe that Ca2+ and Mg2+ induced the soybean roots displaying PCD together and presume what is the mechanism of PCD occurred.
