摘要
收集肝癌等5种消化系癌症患者血清,用免疫电泳法测定血清α1-抗胰蛋酶(α1-AT)水平,结果发现各癌症组血清α1-AT水平均显著高于对照组(P<0.001)。另选择培养两种分化不同的消化系胃癌细胞,分离胞浆及胞核KCl抽提液,用DCC法测定相应性激素受体,结果SGC-7901细胞胞浆及胞核抽提液中ER为30.2和35.7,PR为20.3和22.7fmol/mg蛋白;NKM-45细胞相应ER为24.8和25.1,而PR为16.4和18.6fmol/mg蛋白,证明消化系胃癌细胞为ER、PR阳性,而分化低恶性高转移强的SGC-7901细胞ER、PR含量较高。这对消化系肿瘤的临床诊断有一定价值,也为其内分泌治疗提供证据。
In this experiment we measured the serum alpha-1-antitrypsin level of five sorts of digestive cancers with immunoelectrophoresis methods.The results showed that the serum α1-AT levels of five kinds of digestive cancers were significantly higher than those of mormal control(P<0001).And we chose and cultured two sorts of gastric adenocarcinoma cell lines with different degrees of differentiation,SGC-7901 and NKM-45.We separated the cytosol and nuclear KCl extractable fractions and determined the ER and PR in both fractions of the two cell lines with dextrancoated charcoal method.The results indicated that in the cytosol and nuclear extract the levels of ER were 30.2 and 35.7,the levels of PR were 20.3 and 22.7 fmol/mg protein for SGC-7901 cell,but for NKM-45 cell the levels of ER were 24.8 and 25.1,the levels of PR were 16.4 and 18.6 fmol/mg protein.The results suggested that in a digestive cancer,ER and PR was positive.The higher ER.PR Levels were found in SGC7901 cell line with low differentiation and high tendency of metastasis.So the results had some value for clinical diagnosis of digestive cancers and gave some evidence for endocrine therapy of gastric cancer.
