细胞色素P4501A1、谷胱甘肽S-转移酶基因多态性和急性淋巴细胞白血病的关系研究 被引量:5

摘要 目的探讨广西地区生物代谢酶细胞色素P4501A1(CYP1A1)、谷胱甘肽S-转移酶MI(GSTM1)、谷胱甘肽S-转移酶TI(GSTT1)基因多态性和急性淋巴细胞白血病(ALL)发病的关系。方法采用病例对照研究方法,应用聚合酶链反应-限制性片段长度多态性(PCR—RFLP)技术对88例ALL患者以及120例健康对照者的CYPIA1MSP1多态、GSTM1和GSTT1等基因的多态分布进行分析。结果ALL组的CYP1A1基因MSP1多态纯合子突变型(C型)的频率与对照组差异有显著性(P〈0.05).携带C型的个体患ALL的危险度比杂合子突变型(B型)与野生型(A型)个体的高(OR=1.97,95%CI:1.06~3.64)。ALL组中单一的GETM1或GSTT1缺失型分布频率与对照组相比差异无显著性(P〉0.05)。同时携带CYPIA1C型和GSTM1、GSTT1缺失型的联合基因型个体患ALL的风险增加(OR=2.20,95%CI:1.15~4.22)。结论CYP1A1基因MSP1多态C型与ALL的易感性可能有关;单一的GSTM1或GSTT1缺失型与ALL易感性可能无关。同时携带CYPIA1C型和GSTM1、GSTT1缺失基因型可能是ALL发病的易感因素之一。 Objective To study the relation of metabolic enzymes polymorphism of cytochrome P4501A1 (CYP1A1) and glutathione S- transferase (GST) M1, T1 genotypes and susceptibility to acute lymphoblastic leukemia (ALL). Methods 88 patients with ALL and 120 healthy individuals participated in this casecontrol study. A PCR- RFLP technique was used to detect the polymorphism and frequencies of the genotypes of CYP1A1 MSP1 and GST M1, T1. Results Compared homozygous mutation (type C) of CYP1A1 gene (MSP1 polymorphisms) between group ALL and the control group, there were statistically different ( P 〈 0.05). The individuals with homozygous mutation (type C) enhanced the risk of ALL than those with heterozygote (type B) and wild type (type A) ( OR = 1.97, 95% CI :1.06--3.64). The frequencies of null genotype of GST M1 or GST T1 deletion in group ALL were almost the same as in the control group, there was no statistical difference ( P 〉 0.05). Homozygous mutation of CYP1A1 gene in combination with the null genotype of GST M1, T1 enhanced the risk of ALL ( OR = 2.20, 95 % CI :1.15- 4.22). Conclusion These results indicated that homozygous mutation of CYP1A1 gene might be associated with the susceptibility of ALL. Homozygous mutation of CYP1A1 gene in combination with the null genotypes of GST M1, T1 might serve as a risk factor to the etiology of ALL.
