摘要
观察恶性淋巴瘤中细胞凋亡与细胞增殖的特征,探讨其发生机制。用TUNEL法检测调亡细胞,观察其特征,以每平方毫米面积中的凋亡细胞数作为凋亡指数(apoptoticindex,AI);用枸椽酸-微波-ABC法检测PCNA表达并以PCNA阳性细胞百分率作为增殖指数(PCNAindex,PI)。结果:与反应性增生相比,在大多数淋巴瘤类型中AI显著减小,PI显著增大,而AI/PI在所有淋巴瘤类型中均显著减小。结论:所用检测方法客观而敏感;细胞凋亡减少是恶性淋巴瘤发生的重要原因,特别是小淋巴细胞性、滤泡性、弥漫小裂细胞性淋巴瘤发生的主要原因;AI/PI可能是诊断恶性淋巴瘤、评价其生物学行为的可靠参数。
PURPOSE To observe the characteristics of apoptosis and proliferation in malignant lymphotna and toexplore the mechanism of development of lymphomas.METHODS Apoptotic cells were identified by the terminal deoxynucleotidyl transferase-mediated dUTPnick end labeling (TUNEL) method and the number of apoptotic cells in 1 mm2 of neoplasm was counted asapoptotic index (Al); Using citric acid-microwave-ABC immunohistochemical method, PCNA expressionwas detected and the percentage of positive cells was used as proliferative index (PI).RESULTS Compared with reactive hyperplasia, AI was significantly reduced (P<0. 05 ) and PI was increased (P< 0. 05) in most lymphoma subtypes. The Al/PI ratios in all types was remarkably reduced (P<0. 01).CONCLUSIONS The detective methods are objective and sensitive. Reduced apoptosis is an importantreason in the development of lymphomas and specially is a main mechanism in small lymphocytic, follicular, diffuse small cleaved cell lymphomas. The research provides reliable data for the study of apoptosis induction andgenes regulation in treatment of malignant lyrnphomas.
